高壓氧對慢性疲勞綜合征大鼠影響的實驗研究

【摘要】:慢性疲勞綜合征(Chronic Fatigue Syndrome,CFS)是一組以長期極度疲勞為主要表現(xiàn)的全身性綜合征。隨著現(xiàn)代生活節(jié)奏的加快,慢性疲勞綜合征(chronic fatiguesyndrome,CFS)的發(fā)病率日益增高,嚴重影響了人們生活。我國的CSF發(fā)病率為10%~20%,但增長速度驚人。由于CFS的病因還不明確,因此缺乏特異的動物模型和治療手段。 高壓氧治療可以提高血氧含量,改善細胞代謝,調節(jié)身體各器官功能,可緩解HIV感染和劇烈運動等原因引起的疲勞。 1目的 通過建立CFS動物模型,并予以高壓氧治療,觀察動物的行為學改變以及甲狀腺激素(TSH、T3、T4)、性激素(FSH、LH)、血管內皮素等指標的改變,評價高壓氧治療應用于CFS治療的價值。 2.方法 2.1將SD大鼠隨機分為空白對照組、高壓氧對照組、CFS模型組、CFS高壓氧組及CFS休息組,采用負重力竭游泳的方法建立CFS模型。評價各組大鼠的一般情況、力竭游泳時間、爬桿實驗、曠場實驗的完成情況。 2.2采用放射免疫法檢測血清甲狀腺激素(TSH、T3、T4)水平 2.3采用放射免疫法檢測血管內皮功能:血漿內皮素-1(endothelin-1,ET-1)、一氧化氮(nitric oxide,NO)及血管性假血友病因子(von wille-brand Factor,vWF)的影響。 2.4采用放射免疫法檢測性激素(FSH、LH) 3.結果 3.1CFS模型組大鼠與空白對照組比較,大鼠一般情況變差,體重減輕,力竭游泳時間縮短,爬桿時間縮短,曠場實驗中央格滯留時間明顯延長,跨格次數(shù)、修飾時間及修飾次數(shù)明顯減少(7項指標)。CFS高壓氧組與CFS模型組比較,大鼠的上述7項指標明顯好轉(P0.05);但CFS高壓氧組與空白對照組比較,大鼠7項指標仍恢復不理想(P0.05)。CFS休息組與CFS模型組比較,7項指標無明顯改善(P0.05)。 3.2CFS模型組較空白對照組血清三碘甲腺原氨酸(T3)明顯降低(P0.05);CFS高壓氧組T3水平較CFS模型組有所恢復(P0.05),具有統(tǒng)計學意義;CFS高壓氧組T3較空白對照組、高壓氧對照組仍偏低(P0.05);5組大鼠的TSH、T4水平無明顯差異,P0.05。 3.3CFS模型組較空白對照組血清ET-1、vWF升高(P0.05),而NO降低(P0.05);CFS高壓氧組以上指標較CFS模型組呈反向變化(P0.05),具有統(tǒng)計學意義;CFS高壓氧組以上指標較空白對照組、高壓氧對照組無差異(P0.05)。 3.4CFS高壓氧組促性腺激素(FSH、LH)水平較CFS模型組、CFS休息組有明顯恢復,具有統(tǒng)計學意義。空白對照組與高壓氧對照組激素水平無顯著差異。 4.結論 4.1慢性疲勞綜合征大鼠單純休息不能緩解,高壓氧治療可緩解其癥狀,正常大鼠行高壓氧治療對其行為學沒有明顯影響。 4.2慢性疲勞綜合征大鼠血清T3下降,,經(jīng)高壓氧治療后T3水平較前有所恢復,說明高壓氧對慢性疲勞綜合征有積極作用。 4.3慢性疲勞綜合征大鼠存在血管內皮功能障礙,高壓氧治療能夠改善模型動物的血管內皮功能。 4.4慢性疲勞綜合征大鼠高壓氧治療后促性腺激素(FSH、LH)較前明顯恢復,正常動物行高壓氧治療對促性腺激素(FSH、LH)水平無影響
[Abstract]:Chronic fatigue syndrome (Chronic Fatigue, Syndrome, CFS) is a systemic syndrome in a group of long-term fatigue mainly. With the fast pace of modern life, chronic fatigue syndrome (chronic, fatiguesyndrome, CFS) the incidence is increasing, serious impact on people living in China. The incidence of CSF was 10% to 20%, but the growth rate is amazing. Due to the etiology of CFS is not clear, so the lack of animal model and specific treatment.
Hyperbaric oxygen therapy can improve blood oxygen content, improve cell metabolism, regulate the function of various organs of the body, and relieve the fatigue caused by HIV infection and severe exercise.
1 purposes
By establishing CFS animal model and hyperbaric oxygen therapy, we observed animal behavior changes, TSH T3 (T4), sex hormones (FSH, LH), et et al, and evaluated the value of hyperbaric oxygen therapy in CFS treatment.
2. method
2.1 SD rats were randomly divided into blank control group, hyperbaric oxygen control group, CFS model group, CFS hyperbaric oxygen group and CFS rest group. CFS model was established by weight-bearing exhaustive swimming. The general condition, exhaustive swimming time, pole climbing test and open field experiment were evaluated.
2.2 the levels of serum thyroid hormone (TSH, T3, T4) were detected by radioimmunoassay
2.3, the effects of plasma endothelin -1 (endothelin-1, ET-1), nitric oxide (NO) and von Willebrand factor (von wille-brand Factor, vWF) were detected by radioimmunoassay.
2.4 FSH (LH) was detected by radioimmunoassay.
3. results
With the blank group rats 3.1CFS model control group, rats in general variation, weight loss, exhaustive swimming time, climbing pole time, residence time of the open field test in central square was prolonged, the grid crossing frequency, modification time and modification times were significantly reduced (7 indicators).CFS hyperbaric oxygen group and CFS compared to the model group, the rats of the 7 indicators improved significantly (P0.05); but CFS hyperbaric oxygen group compared with the control group, 7 rats were still not ideal recovery index (P0.05).CFS rest group comparing with CFS model group, 7 indexes had no obvious improvement (P0.05).
The 3.2CFS model group compared with blank control group, three serum triiodothyronine (T3) decreased significantly (P0.05); CFS hyperbaric oxygen group T3 levels compared with CFS group (P0.05) has been restored, with statistical significance; CFS T3 in HBO group than the control group, hyperbaric oxygen group (P0.05) is still low; 5 rats in group TSH, no significant differences in the level of T4, P0.05.
The 3.3CFS model group compared with blank control group, serum ET-1 and vWF increased (P0.05), and NO decreased (P0.05); CFS hyperbaric oxygen group compared with CFS group the above indexes showed the reverse changes (P0.05), with statistical significance; CFS index above the hyperbaric oxygen group compared with blank control group, no difference in hyperbaric oxygen group (P0.05).
The level of FSH (LH) in 3.4CFS hyperbaric oxygen group was significantly higher than that in CFS model group and CFS rest group, and there was statistical significance. There was no significant difference in hormone level between blank control group and hyperbaric oxygen control group.
4. conclusion
4.1, chronic fatigue syndrome rats can not alleviate rest alone. Hyperbaric oxygen therapy can relieve their symptoms.
4.2 the serum T3 of rats with chronic fatigue syndrome decreased and the level of T3 recovered after hyperbaric oxygen therapy. It indicated that hyperbaric oxygen had a positive effect on chronic fatigue syndrome.
4.3 the rats with chronic fatigue syndrome have vascular endothelial dysfunction, and hyperbaric oxygen therapy can improve the vascular endothelial function of model animals.
4.4, FSH, LH in chronic fatigue syndrome rats were significantly restored after hyperbaric oxygen treatment. No significant difference was found in the level of gonadotrophin (FSH, LH) in normal animals.
