抑郁癥大鼠海馬體積異常的形態(tài)學(xué)探討

【摘要】:目的觀察抑郁癥模型大鼠海馬神經(jīng)元形態(tài)結(jié)構(gòu)改變及凋亡、自噬的變化,探討抑郁癥海馬體積異常的機(jī)制。方法選用雄性成年SD大鼠,隨機(jī)分為正常對(duì)照組和模型組,通過(guò)給予不可預(yù)見(jiàn)慢性溫和應(yīng)激建立抑郁癥模型;采用尼氏染色觀察海馬神經(jīng)元形態(tài)變化,流式細(xì)胞術(shù)檢測(cè)海馬神經(jīng)元凋亡,Western blotting檢測(cè)自噬相關(guān)蛋白LC-3和Beclin 1。結(jié)果與對(duì)照組比較,模型組海馬體積萎縮,神經(jīng)元數(shù)量減少,細(xì)胞凋亡率增高(P0.05)。與對(duì)照組相比,模型組LC-3Ⅱ蛋白和LC-3Ⅱ/LC-3Ⅰ比值增高,Beclin 1相對(duì)表達(dá)增高(P0.05)。結(jié)論抑郁癥大鼠海馬凋亡和自噬增強(qiáng),可能是海馬體積異常的原因之一。
[Abstract]:Objective to observe the morphological changes, apoptosis and autophagy of hippocampal neurons in rats with depression, and to explore the mechanism of abnormal hippocampal volume. Methods male adult Sprague-Dawley rats were randomly divided into normal control group and model group. Depression model was established by giving unpredictable chronic mild stress, morphological changes of hippocampal neurons were observed by Nissl staining, apoptosis of hippocampal neurons was detected by flow cytometry, autophagy associated protein LC-3 and Beclin 1 were detected by flow cytometry.
