【摘要】：目的探討青春期亞慢性地卓西平(MK-801)注射對(duì)小鼠成年期前脈沖抑制(PPI)和糖耐量的影響。方法 20只青春期小鼠隨機(jī)分為對(duì)照組(n=10)和模型組(n=10),兩組小鼠出生后30~43 d分別接受0.9%生理鹽水(4 mL/kg)和MK-801(0.6 mg/kg)注射,1次/d,持續(xù)2周。每周記錄攝食和體質(zhì)量,并于小鼠成年期(出生后70~80 d之間)進(jìn)行曠場(chǎng)試驗(yàn)、PPI測(cè)試和葡萄糖耐量試驗(yàn)(GTT)。結(jié)果模型組小鼠在79 dB強(qiáng)度的前脈沖刺激時(shí),PPI顯著低于對(duì)照組(P0.05),表明MK-801成功誘導(dǎo)小鼠精神分裂樣行為。模型組小鼠體質(zhì)量也顯著低于對(duì)照組(P0.05)。GTT結(jié)果顯示:模型組小鼠注射葡萄糖后90 min和120 min的血糖水平顯著高于對(duì)照組(P0.001和P0.01),表明MK-801誘導(dǎo)的精神分裂樣小鼠模型出現(xiàn)糖耐量異常。兩組小鼠的攝食量和曠場(chǎng)試驗(yàn)中的自發(fā)活動(dòng)量差異均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論青春期亞慢性注射MK-801可誘導(dǎo)小鼠成年期PPI損害和糖耐量異常。
[Abstract]:Objective to investigate the effects of subchronic Dizoxipine MK-801) injection on preadulthood pulse inhibition of PPI) and glucose tolerance in mice. Methods Twenty puberty mice were randomly divided into two groups: control group (n = 10) and model group (n = 30, 43 d after birth). Don't take 0.9% saline (4 mL / kg) and MK-801(0.6 mg / kg once a day for 2 weeks. Record your food intake and body mass weekly. Open field test (PPI) and glucose tolerance test (GTT) were performed in adult mice (between 70 and 80 days after birth). Results the PPI of the model group was significantly lower than that of the control group at 79 dB intensity prepulse stimulation, indicating that MK-801 was successfully induced small. The body mass of mice in the model group was significantly lower than that in the control group (P 0.05). The results showed that the blood glucose levels in the model group at 90 min and 120 min after glucose injection were significantly higher than those in the control group (P 0.001 and P 0.01), indicating that the psychosis induced by MK-801 in the model group was significantly higher than that in the control group (P 0.001 and P 0.01). There was no significant difference between the two groups in food intake and spontaneous activity in open field test (P 0.05). Conclusion Subchronic injection of MK-801 in puberty can induce PPI damage and impaired glucose tolerance in adult mice.
