【摘要】：目的探討海馬齒狀回(DG)的GABAB受體是否參與血管性癡呆(VD)大鼠空間學(xué)習(xí)記憶損害。方法 SD雄性大鼠24只,隨機(jī)分為假手術(shù)組、VD組及GABAB受體阻斷劑沙克洛芬組,雙側(cè)頸總動(dòng)脈永久性結(jié)扎法制備VD大鼠模型。利用Morris水迷宮檢測(cè)大鼠的空間學(xué)習(xí)記憶能力;腦部微量透析法和高效液相色譜法測(cè)定DG區(qū)γ-氨基丁酸(GABA)含量;免疫組織化學(xué)法觀察DG區(qū)GABAB受體表達(dá);用腦部微量注射法往DG區(qū)注射GABAB受體阻斷劑沙克洛芬,觀察其對(duì)VD模型大鼠空間學(xué)習(xí)記憶的影響。結(jié)果 VD大鼠海馬DG區(qū)GABA含量明顯增加,而GABAB受體表達(dá)減少(均為P0.05)。VD大鼠的空間學(xué)習(xí)記憶能力顯著下降,而訓(xùn)練前向DG區(qū)微量注射GABAB受體阻斷劑可改善VD大鼠的空間學(xué)習(xí)記憶損害。結(jié)論 VD模型大鼠海馬DG內(nèi)增加的GABA通過(guò)GABAB受體參與其空間學(xué)習(xí)記憶的損害。
[Abstract]:Objective to investigate the dentate gyrus (DG) whether the GABAB receptor is involved in vascular dementia (VD) rats learning and memory impairment. Methods 24 male SD rats were randomly divided into sham operation group, VD group and GABAB receptor antagonist group Shah Chloe Finn, permanent bilateral common carotid artery ligation model was prepared by the method of VD the ability of learning and memory in rats. Morris water maze test in rats; Determination of DG gamma aminobutyric acid microdialysis and high performance liquid chromatography (GABA) content; immunohistochemical method to observe the expression of DG GABAB receptor in the brain; microinjection into DG microinjection of GABAB receptor antagonist Shah Chloe Finn, observation the changes of learning and memory in VD rats. The content of GABA in hippocampus DG area of VD rats increased significantly, and reduce the expression of GABAB receptor (P0.05).VD spatial learning and memory ability in rats decreased significantly, while training the DG prior to microinjection GABAB receptor antagonist can improve the spatial learning and memory impairment of VD rats. Conclusion GABA increased in hippocampus DG of VD rats is involved in spatial learning and memory impairment through GABAB receptor.
