雙七他克林對青光眼模型大鼠視神經(jīng)的保護作用

目的:觀察雙七他克林對青光眼模型大鼠的視神經(jīng)保護作用。方法:選取雄性成年SD大鼠24只,隨機分成對照組、手術(shù)組、雙七他克林組和美金剛胺組,通過右眼鞏膜上靜脈注射高滲鹽水(對照組大鼠右眼僅進行上鞏膜靜脈穿刺)制作青光眼模型后,對照組和手術(shù)組大鼠給予正常飲水,雙七他克林組和美金剛胺組大鼠飲用水中每天分別添加0.5mg/kg雙七他克林和5mg/kg美金剛胺,持續(xù)5wk。定期測量眼壓,造模后5wk處死大鼠,取右側(cè)視網(wǎng)膜進行免疫熒光染色,檢測視網(wǎng)膜神經(jīng)節(jié)細胞數(shù)目和視網(wǎng)膜神經(jīng)纖維層厚度。結(jié)果:造模后模型大鼠右眼眼壓均顯著升高,至造模后1mo,均高于28mmHg。手術(shù)組大鼠右眼視網(wǎng)膜神經(jīng)節(jié)細胞數(shù)較對照組明顯減少(79.83±9.58個vs 119.50±8.26個,P<0.01),神經(jīng)纖維層厚度明顯變。6.64±0.50μm vs 13.40±0.60μm,P<0.01),而雙七他克林(109.00±7.04個)和美金剛胺(107.33±8.57個)能夠抑制青光眼所致的視網(wǎng)膜神經(jīng)節(jié)細胞減少,且雙七他克林能夠抑制青光眼所致的視網(wǎng)膜神經(jīng)纖維層萎縮(12.26±0.78μm)。結(jié)論:雙七他克...
