海馬TLR4及TNF-α參與CCI大鼠神經(jīng)痛的初步研究

目的:1.闡明海馬TLR4(Toll like receptor 4,Toll樣受體4)及TNF-α(Tumor Necrosis Factor-alpha,腫瘤壞死因子-α)表達(dá)是否參與CCI(chronic constriction injury of sciatic nerve,坐骨神經(jīng)慢性縮窄性損傷)大鼠神經(jīng)痛。2.闡明雙側(cè)海馬注射米諾環(huán)素或miRNA-181c激動(dòng)劑能否通過(guò)下調(diào)海馬TLR4及TNF-α表達(dá)緩解CCI大鼠神經(jīng)痛。方法:選用56周齡SD雄性大鼠,均行海馬CA1區(qū)置管。1.SD大鼠,隨機(jī)分成3組(n=8):(1)假手術(shù)組(Sham組)、(2)CCI+溶劑對(duì)照組(CCI+0.01 mol/L PBS)、(3)CCI+米諾環(huán)素組(CCI+2μg/μL米諾環(huán)素)。CCI+米諾環(huán)素組雙側(cè)海馬CA1區(qū)注射米諾環(huán)素(2μg/μL,2次/日,持續(xù)7 d)。假手術(shù)組及CCI+溶劑對(duì)照組雙側(cè)海馬CA1區(qū)注射等體積的0.01M PBS(持續(xù)7 d)。于術(shù)前1 d,術(shù)后第1、3、5、7 d測(cè)定給藥1 h后的MWT(mechanical withdraw thresh...
