缺氧預(yù)處理對豬缺氧復(fù)氧冠狀動脈內(nèi)皮舒張功能的影響

【摘要】:目的:探討缺氧預(yù)處理對豬缺氧復(fù)氧冠狀動脈內(nèi)皮源性超極化因子(EDHF)介導(dǎo)的內(nèi)皮舒張功能的影響及其機(jī)制。 方法:隨機(jī)選取新鮮豬心9個(gè),每只取其心外膜下冠狀動脈前降支中下1/3切成4段,長2m,隨機(jī)置入如下4組的環(huán)境中。對照組:冠狀動脈血管環(huán)未經(jīng)過缺氧/復(fù)氧處理及缺氧預(yù)處理,直接在37℃有氧條件下用KH液孵育60mmin,再平衡30min;實(shí)驗(yàn)組A:在4℃條件下用KH液缺氧浸泡60min后復(fù)氧30min,再按對照組的方法進(jìn)行處理;實(shí)驗(yàn)組B:在4℃條件下用KH液缺氧浸泡5min后復(fù)氧10min,再按實(shí)驗(yàn)組A的方法進(jìn)行處理;實(shí)驗(yàn)組C:在37℃有氧條件下含5-羥基癸酸甘油酯(10μmol/L)的溶液浸泡25min,再按實(shí)驗(yàn)組B的方法進(jìn)行處理。然后采用器官槽法檢測血管環(huán)在消炎痛(7μmol/L)、N-硝基-L-精氨酸(300μ/L)及氧合血紅蛋白(20μmol/L)作用下,前列腺素F2α引發(fā)的血管收縮反應(yīng),及不同緩激肽濃度下EDHF引發(fā)的血管舒張反應(yīng)。 結(jié)果:與對照組相比,內(nèi)皮源性超極化因子引發(fā)的血管環(huán)最大舒張反應(yīng)變化,實(shí)驗(yàn)組A、C均明顯降低(P0.001),而實(shí)驗(yàn)組B變化不明顯(P0.05)。 結(jié)論:①缺氧復(fù)氧對冠狀動脈內(nèi)皮源性超極化因子所介導(dǎo)的內(nèi)皮依賴性舒張功能有損害作用;②缺氧預(yù)處理對冠狀動脈內(nèi)皮源性超極化因子所介導(dǎo)的內(nèi)皮依賴性舒張功能有保護(hù)作用;③缺氧預(yù)處理能選擇性開放線粒體ATP敏感的鉀離子通道(mitochondria ATP-sensitive K+channel, mKATP)可能是這一保護(hù)作用的主要原因。
