【摘要】：目的:探討有絲分裂相關(guān)激酶Plk1磷酸化修飾Tara蛋白在調(diào)控HeLa細(xì)胞胞質(zhì)分裂及增殖中的作用。方法:運(yùn)用酵母雙雜交、免疫共沉淀、體外沉降實(shí)驗(yàn)確定Plk1與Tara兩種蛋白是否能夠相互結(jié)合;采用體內(nèi)外磷酸化實(shí)驗(yàn)明確Plk1對(duì)Tara蛋白的磷酸化修飾;采用免疫熒光實(shí)驗(yàn)及流式細(xì)胞術(shù)檢測(cè)Tara蛋白的磷酸化對(duì)HeLa細(xì)胞胞質(zhì)分裂及細(xì)胞增殖的影響。結(jié)果:Plk1與Tara在體內(nèi)外均能結(jié)合;Tara蛋白在體內(nèi)外均能夠被Plk1磷酸化修飾;過(guò)量表達(dá)Tara蛋白的磷酸化突變體對(duì)HeLa細(xì)胞的胞質(zhì)分裂有明顯的抑制作用,細(xì)胞增殖明顯受阻。結(jié)論:Plk1能夠與Tara結(jié)合,并通過(guò)磷酸化修飾Tara蛋白阻礙細(xì)胞胞質(zhì)分裂進(jìn)程及抑制細(xì)胞增殖。
[Abstract]:Aim: to investigate the role of mitotic kinase Plk1 phosphorylation modified Tara protein in regulating cytoplasmic division and proliferation of HeLa cells. Methods: yeast two-hybrid, immunoprecipitation, in vitro sedimentation assay were used to determine whether Plk1 and Tara could bind to each other, and in vitro and in vivo phosphorylation experiments were used to determine the phosphorylation modification of Tara protein by Plk1. The effects of phosphorylation of Tara protein on cytokinesis and proliferation of HeLa cells were detected by immunofluorescence assay and flow cytometry. Results: Plk1 and Tara could bind in vivo and in vitro, Tara protein could be modified by Plk1 phosphorylation in vitro and in vivo, and the phosphorylated mutants which overexpressed Tara protein could inhibit the cytokinesis of HeLa cells and inhibit the proliferation of HeLa cells. Conclusion: Plk1 can bind to Tara and modify Tara protein by phosphorylation to block the process of cytokinesis and inhibit cell proliferation.
