激光致猴脈絡(luò)膜新生血管模型的應(yīng)用

【摘要】:目的通過激光誘導(dǎo)猴眼底黃斑區(qū)脈絡(luò)膜新生血管(CNV)滲漏和生長,建立與人脈絡(luò)膜新生血管類似的動(dòng)物模型,對(duì)擬用于治療脈絡(luò)膜新生血管相關(guān)疾病藥物的藥效學(xué)進(jìn)行評(píng)價(jià)。方法采用眼科激光光凝儀圍繞猴黃斑中心凹進(jìn)行光凝造模,光凝后2~3周,進(jìn)行熒光素眼底血管造影檢查,篩選出造模成功的動(dòng)物,根據(jù)造模效果分為模型對(duì)照組、陽性對(duì)照組、藥物低、中、高劑量組。分別于給藥前、給藥后14和28 d進(jìn)行眼底彩色照相、熒光素眼底血管造影(FFA)、光學(xué)相干斷層掃描(OCT)檢查,給藥后29 d解剖動(dòng)物進(jìn)行眼球組織病理學(xué)檢查。結(jié)果陽性對(duì)照組、藥物低、中、高劑量組給藥后熒光素滲漏面積明顯減少,激光斑病損處視網(wǎng)膜厚度明顯降低。組織學(xué)切片觀察到模型對(duì)照組CNV視網(wǎng)膜神經(jīng)上皮脫離及神經(jīng)上皮下水腫,陽性對(duì)照組、藥物低、中、高劑量組眼球基本未見異常組織學(xué)改變。結(jié)論該藥物在各劑量下均對(duì)激光所致的猴脈絡(luò)膜新生血管具有較明顯的抑制作用。激光誘導(dǎo)的猴脈絡(luò)膜新生血管模型穩(wěn)定持久,是研究治療與脈絡(luò)膜新生血管相關(guān)眼底病藥物的理想動(dòng)物模型。
[Abstract]:Objective to establish an animal model of choroidal neovascularization similar to human choroidal neovascularization by laser induced choroidal neovascularization (CNV) leakage and growth in the macular region of monkey eye, and to evaluate the pharmacodynamics of the drugs proposed for the treatment of choroidal neovascularization related diseases. Methods the ophthalmic laser photocoagulation instrument was used to model the macular fovea of monkey. After 2 weeks of photocoagulation, fundus fluorescein angiography was performed. The successful animals were selected and divided into model control group according to the model effect. Positive control group, low, medium and high dose group. Fundus color photography was performed before administration, 14 d and 28 d after administration, fundus fluorescein angiography (FFA),) optical coherence tomography (OCT) and histopathological examination of eyeball were performed 29 days after administration. Results in the positive control group, the area of fluorescein leakage was significantly decreased and the retinal thickness of laser spot lesion was significantly decreased in the low, middle and high dose groups. Histopathological examination showed that the retinal neuroepithelial detachment and subcutaneous edema of CNV were observed in the model control group, but no abnormal histological changes were observed in the low, medium and high dose groups of the positive control group. Conclusion the drug has obvious inhibitory effect on the choroidal neovascularization induced by laser at various doses. The laser-induced choroidal neovascularization model is an ideal animal model for the treatment of choroidal angiogenic fundus disease.
