間日瘧原蟲環(huán)子孢子蛋白PCR-RFLP多態(tài)性分析

【摘要】:采集2011年河南省間日瘧原蟲陽(yáng)性患者血樣,用限制性片段長(zhǎng)度多態(tài)性聚合酶鏈反應(yīng)(PCR-RFLP)法分析環(huán)子孢子蛋白(circumsporozoite protein,CSP)基因型,并結(jié)合流行病學(xué)資料進(jìn)行初步種群分布分析。共采集間日瘧患者血樣37份,根據(jù)流行病學(xué)調(diào)查確定河南當(dāng)?shù)馗腥静±獦?6份,輸入性病例血樣11份。37份血樣均擴(kuò)增出1條約750 bp的條帶。PCR-RFLP結(jié)果顯示,CSP基因PV-Ⅱ型1份,占2.7%,PV-Ⅰ型36份,占97.3%。26份當(dāng)?shù)夭±獦覥SP基因均為PV-Ⅰ型,其中PV-Ⅰ-a型占42.3%(11/26),PV-Ⅰ-b型占57.7%(15/26);11份輸入病例血樣中,PV-Ⅰ-a型占90.9%(10/11),PV-Ⅱ型占9.1%(1/11),未見PV-Ⅰ-b型。提示河南省當(dāng)?shù)亓餍械拈g日瘧病例中,CSP基因PV-Ⅰ-b型略多于PV-Ⅰ-a型;輸入性間日瘧病例中PV-Ⅰ-a型占優(yōu)勢(shì)。
[Abstract]:Blood samples of Plasmodium vivax positive patients in Henan Province were collected in 2011. The genotypes of circumsporozoite protein were analyzed by restriction fragment length polymorphism polymerase chain reaction (PCR-RFLP), and the preliminary population distribution was analyzed with epidemiological data. A total of 37 blood samples were collected from vivax malaria patients. 26 blood samples were collected from local infection cases in Henan Province, and 11 blood samples from imported cases were collected.
