【摘要】：目的:探討內(nèi)毒素脂多糖(Lipopolysaccharide,LPS)對(duì)大鼠星形膠質(zhì)細(xì)胞熱休克蛋白A12B(Heat shock proteins A 12B,HSPA12B)表達(dá)變化和細(xì)胞定位的影響。方法:用不同濃度(0.01、0.1、1、10μg/ml)和同一濃度(1μg/ml)不同時(shí)間(1、2、4、6、8、12、24小時(shí))的LPS刺激星形膠質(zhì)細(xì)胞,分別用RT-PCR和Western blot法檢測(cè)星形膠質(zhì)細(xì)胞HSPA12B的mRNA和蛋白質(zhì)水平表達(dá)情況,間接免疫熒光雙標(biāo)法檢測(cè)HSPA12B在星形膠質(zhì)細(xì)胞中的細(xì)胞定位。結(jié)果:用LPS 1μg/ml刺激1小時(shí)后,HSPA12B mRNA的表達(dá)增加,4小時(shí)表達(dá)活性最高。HSPA12B蛋白水平表達(dá)變化與mRNA水平變化相一致。免疫細(xì)胞化學(xué)證明LPS誘導(dǎo)星形膠質(zhì)細(xì)胞HSPA12B的表達(dá)定位在胞核。結(jié)論:LPS可誘導(dǎo)星形膠質(zhì)細(xì)胞HSPA12B表達(dá)上調(diào)。HSPA12B可作為一種炎癥反應(yīng)蛋白參與炎癥反應(yīng)過(guò)程。
[Abstract]:Aim: to investigate the effect of lipopolysaccharide (LPS) on the expression of heat shock protein (A12B(Heat shock proteins A 12B) and its cellular localization in rat astrocytes. Methods: the astrocytes were stimulated with LPS at different concentrations of 0. 01 ~ 0. 1 ~ 1 ~ (1 渭 g / ml) and 1 渭 g / ml of the same concentration of 1 渭 g / ml). The expression of mRNA and protein in astrocytes was detected by RT-PCR and Western blot methods. The localization of HSPA12B in astrocytes was detected by indirect immunofluorescence double labeling method. Results: after 1 hour stimulation with LPS 1 渭 g/ml, the expression of HSPA12B mRNA was increased. The expression of HSPA12B protein was the highest at 4 hours. The change of HSPA12B protein level was consistent with the change of mRNA level. Immunocytochemistry showed that the expression of HSPA12B in astrocytes induced by LPS was localized in the nucleus. Conclusion the up-regulation of HSPA12B expression and HSPA12B may be involved in the process of inflammation as a kind of inflammatory response protein.
