【摘要】：目的:觀察血管生成素4(Ang-4)對(duì)脂多糖(LPS)誘導(dǎo)的人臍靜脈內(nèi)皮細(xì)胞(HUVECs)損傷的影響,并探討其可能的作用機(jī)制。方法:EnVision法免疫組化鑒定HUVECs。LPS誘導(dǎo)細(xì)胞損傷,不同劑量的Ang-4干預(yù)。顯微鏡下觀察細(xì)胞形態(tài),MTT法檢測(cè)細(xì)胞活力,ELISA法檢測(cè)von Willebrand因子(vWF)和腫瘤壞死因子(TNF-α)含量,real-time PCR檢測(cè)Toll樣受體4(TLR4)、核因子κB(NF-κB)p65和TNF-αmRNA含量變化。結(jié)果:免疫組化示胞漿內(nèi)人Ⅷ因子相關(guān)抗原呈陽性;與正常組比,LPS組細(xì)胞增殖活力降低(P0.01),vWF及TNF-α分泌增多(P0.01),且TLR4、NF-κB p65和TNF-αmRNA表達(dá)升高(P0.01);Ang-4(100μg/L)組可恢復(fù)細(xì)胞活力,降低vWF及TNF-α含量(P0.01),抑制LPS所致TLR4、NF-κB p65和TNF-αmRNA的表達(dá)(P0.01)。結(jié)論:Ang-4可拮抗LPS誘導(dǎo)的HUVECs損傷,這可能與抑制TLR4-NF-κB p65-TNF-α信號(hào)通路的活化有關(guān)。
[Abstract]:Aim: to observe the effect of Ang-4 on the injury of HUVECs induced by lipopolysaccharide (LPS) in human umbilical vein endothelial cells (HUVECs) and to explore its possible mechanism. Methods the cell injury induced by HUVECs.LPS was identified by immunohistochemistry with different doses of Ang-4. The contents of von Willebrand factor vWFand TNF- 偽 were detected by MTT assay under microscope.
