舒心湯對(duì)抑郁模型大鼠治療作用的實(shí)驗(yàn)研究

【摘要】:目的通過舒心湯干預(yù)孤養(yǎng)與慢性溫和不可預(yù)見性應(yīng)激(CUMS)抑郁模型大鼠的行為學(xué)及下丘腦-垂體-腎上腺軸(HPAA)和炎性因子白介素1β(IL-1β)、白介素6(IL-6)的結(jié)果差異,觀察舒心湯對(duì)抑郁模型大鼠的療效。根據(jù)中醫(yī)調(diào)理臟氣理論,探討舒心湯的抗抑郁機(jī)制,為臨床應(yīng)用舒心湯治療抑郁癥提供新的思路。方法以孤養(yǎng)結(jié)合CUMS方法制造抑郁大鼠模型,分別于實(shí)驗(yàn)前和實(shí)驗(yàn)后測(cè)定大鼠的體重、糖水消耗率、敞箱實(shí)驗(yàn)得分、強(qiáng)迫游泳不動(dòng)時(shí)間的變化結(jié)果,其中體重情況每周測(cè)定一次,據(jù)此評(píng)定舒心湯對(duì)孤養(yǎng)結(jié)合CUMS抑郁模型大鼠行為學(xué)的改變情況。按照ELISA試劑盒說明書分別測(cè)量血清促腎上腺皮質(zhì)激素(ACTH)、皮質(zhì)醇(CORT)、白介素6(IL-6)、白介素-1β(IL-1β)水平。RT-PCR檢測(cè)下丘腦內(nèi)CRHm RNA及海馬內(nèi)GRm RNA的表達(dá)。實(shí)驗(yàn)數(shù)據(jù)以(?)±s表示,組間比較應(yīng)用單因素方差分析進(jìn)行統(tǒng)計(jì),以P0.05為有差異性,P0.01為有顯著差異。結(jié)果1 28天的造模應(yīng)激之后,與空白組對(duì)比,模型組的行為學(xué)指標(biāo)均明顯降低(P0.05或P0.01),模型組大鼠的血清ACTH、CORT、IL-1β、IL-6及下丘腦CRHm RNA水平明顯升高(P0.05或P0.01),海馬GRm RNA表達(dá)明顯下降(P0.05或P0.01),結(jié)果有(明顯)差異性。2 28天藥物干預(yù)之后,與模型組對(duì)比,舒心湯組、舍曲林組及聯(lián)合用藥組的行為學(xué)指標(biāo)明顯上升(P0.05或P0.01),血清ACTH、CORT、IL-1β、IL-6及下丘腦CRHm RNA水平明顯下降(P0.05或P0.01),海馬GRm RNA表達(dá)明顯上升(P0.05或P0.01),結(jié)果有(明顯)差異性。結(jié)論1中藥復(fù)方制劑舒心湯能有效改善孤養(yǎng)結(jié)合CUMS抑郁模型大鼠的行為學(xué)指標(biāo),即加速體重的增長,增加糖水消耗率,增加敞箱實(shí)驗(yàn)垂直和水平運(yùn)動(dòng)得分,減少強(qiáng)迫游泳大鼠不動(dòng)時(shí)間。2中藥復(fù)方制劑舒心湯能調(diào)節(jié)抑郁模型大鼠的HPA軸及炎性因子功能,即降低抑郁大鼠血清中ACTH、CORT、IL-1β、IL-6的水平及抑郁大鼠下丘腦CRH的基因表達(dá),提高抑郁大鼠海馬GR的基因表達(dá)。3舒心湯的抗抑郁機(jī)制可能與干預(yù)神經(jīng)內(nèi)分泌免疫系統(tǒng)的HPA軸及炎性因子有關(guān)。
[Abstract]:Objective to study the behavior and the difference of hypothalamus-pituitary-adrenal axis (HPAA) and inflammatory factor IL-1 尾 (IL-1 尾, interleukin-6) in rats with depression induced by Shuxin decoction and chronic mild and unpredictable stress (CUMS).To observe the effect of Shuxin decoction on depression model rats.According to the theory of regulating visceral qi in TCM, the antidepressant mechanism of Shuxin decoction is discussed, which provides a new idea for clinical application of Shuxin decoction in the treatment of depression.Methods the depressive rat model was established by the method of solitary raising and CUMS. The weight, the consumption rate of sugar water, the score of open box test and the time of forced swimming were measured before and after the experiment, in which the body weight was measured once a week.To evaluate the effect of Shuxin decoction on the behavior of rats with solitary care combined with CUMS depression.The expression of CRHm RNA in hypothalamus and GRm RNA in hippocampus were detected by reverse transcription-polymerase chain reaction (RT-PCR) according to the instructions of ELISA kit.The experimental data were expressed as) 鹵s, and the statistical results were analyzed by univariate analysis of variance (ANOVA). There was significant difference between the two groups with P05 as the difference (P0.01).Results after 28 days of stress, the model was compared with the control group.The behavioral indexes of the model group were significantly lower than those of the control group (P0.05 or P0.01). The serum levels of IL-1 尾 -IL-6 and CRHm RNA in the hypothalamus of rats in the model group were significantly higher than those in the control group (P 0.05 or P 0.01), and the expression of GRm RNA in the hippocampus decreased significantly (P 0.05 or P 0.01). The results showed that there were significant differences between the two groups after the drug intervention for 2.28 days.Compared with the model group, the behavioral indexes of Shuxin decoction group, sertraline group and combination group were significantly increased (P 0.05 or P 0.01), the serum levels of ACTH cortisone IL-1 尾 IL-6 and hypothalamus CRHm RNA were significantly decreased (P0.05 or P0.01), and the expression of GRm RNA in hippocampus was significantly increased (P 0.05 or P 0.01).Conclusion (1) Shuxin decoction, a compound preparation of Chinese medicine, can effectively improve the behavioral indexes of rats with solitary rearing combined with CUMS depression model, that is, accelerate the growth of body weight, increase the consumption rate of sugar water, and increase the scores of vertical and horizontal movement in open box experiment.To reduce the immobility time of forced swimming rats. 2. Shuxin decoction can regulate the function of HPA axis and inflammatory factors in depressive rats, that is, it can reduce the level of serum ACTHCORTT IL-1 尾 and IL-6 and the gene expression of CRH in hypothalamus of depressive rats.The antidepressant mechanism of Shuxin decoction may be related to the intervention of HPA axis and inflammatory factors in neuroendocrine immune system.
