甲基對(duì)硫磷對(duì)小鼠黃體細(xì)胞的細(xì)胞間隙連接通訊和孕酮分泌的影響

【摘要】:目的探索甲基對(duì)硫磷(methyl parathion,MP)對(duì)小鼠黃體細(xì)胞的細(xì)胞間隙連接通訊(gap junctional intercellular communication,GJIC)和孕酮分泌的影響。方法體外培養(yǎng)5周齡SPF級(jí)BALB/c小鼠黃體細(xì)胞,分別設(shè)MP暴露組(0、0.05、0.1、0.2、0.4、0.8、1.6 mmol/L的MP處理3 h)和GJIC促進(jìn)劑——1μmol/L異丙腎上腺素預(yù)處理組及蛋白激酶A(PKA)專一抑制劑——10μmol/L H89預(yù)處理組與蛋白激酶C(PKC)專一抑制劑——10μmol/L恩扎妥林預(yù)處理組(預(yù)處理5 min,0.8 mmol/L MP處理3 h)。采用ELISA法檢測(cè)培養(yǎng)液中孕酮含量,劃痕標(biāo)記染料轉(zhuǎn)移法測(cè)定貼壁細(xì)胞的GJIC。結(jié)果高濃度(0.2~1.6 mmol/L)MP可明顯抑制小鼠黃體細(xì)胞的GJIC和孕酮分泌(P0.05);異丙腎上腺素、H89和恩扎妥林均能緩解MP對(duì)體外小鼠黃體細(xì)胞的GJIC和孕酮分泌的抑制作用(P0.05)。結(jié)論 MP可通過抑制小鼠黃體細(xì)胞的GJIC導(dǎo)致孕酮分泌受抑制,而PKA與PKC參與該過程。
[Abstract]:Objective to investigate the effects of methyl parathionine (MPP) on gap junctional intercellular communication and progesterone secretion in mouse luteal cells. Methods SPF grade BALB/c mice luteal cells of 5 weeks old were cultured in vitro. The MP exposure group was divided into two groups: 1 渭 mol/L isoproterenol preconditioning group and 10 渭 mol/L H89 preconditioning group and 10 渭 mol/L Enzaturin preconditioning group (10 渭 mol/L H89 preconditioning group) and 10 渭 mol/L Enzaturin preconditioning group (10 渭 mol/L) respectively after 3 hours of MP treatment with 0. 05 mol/L 0. 1 and 0. 4 and 0. 8 mmol/L) and 1 渭 mol/L isoproterenol preconditioning group and 10 渭 mol/L H89 preconditioning group (10 渭 mol/L Enzaturin preconditioning group, respectively). The concentration of progesterone in the culture medium was determined by ELISA method. The GJICs of adherent cells were determined by scratch labeling dye transfer method. Results the GJIC and progesterone secretion of mouse luteal cells were significantly inhibited by high concentration of 0.2N 1.6 mmol/L)MP, and isoproterenol H89 and Enzarytoin could alleviate the effect of MP on mouse luteal cells in vitro. Conclusion MP can inhibit progesterone secretion by inhibiting GJIC of mouse luteal cells. PKA and PKC participate in the process.
