【摘要】：目的：探討甘露消毒丹及其殘方對(duì)EV71感染模型小鼠機(jī)體免疫炎癥損傷的免疫調(diào)節(jié)作用,并進(jìn)一步認(rèn)識(shí)甘露消毒丹配伍的科學(xué)內(nèi)涵。 方法：①文獻(xiàn)研究：從甘露消毒丹方源、臨床運(yùn)用情況、抗病毒物質(zhì)基礎(chǔ)等方面探討甘露消毒丹治手足口病的有效性。②實(shí)驗(yàn)研究：7日齡ICR小鼠分為空白組、模型組、全方組、芳?xì)埛浇M、清殘方組、利殘方組等6組。在高溫、高濕環(huán)境中腹腔注射造EV71病毒造模并使用相應(yīng)藥物干預(yù)治療。用Elisa法檢測(cè)小鼠靶組織中IL-10、IFN-γ、TNF-α等細(xì)胞因子變化；用流式細(xì)胞術(shù)檢測(cè)小鼠脾臟淋巴細(xì)胞亞群；用Western blot法檢測(cè)靶組織中IRF3蛋白表達(dá)。 結(jié)果：①從文獻(xiàn)研究來看,甘露消毒丹為葉天士所創(chuàng),首載于《續(xù)名醫(yī)類案》具有治療EV71型手足口病的臨床基礎(chǔ)。②EV71感染小鼠后第5天觀測(cè)臨床表現(xiàn),檢測(cè)后肢肌肉組織形態(tài)及是否表達(dá)了EV71病毒,均符合造模標(biāo)準(zhǔn),確認(rèn)造模成功。③甘露消毒丹及其殘方能調(diào)節(jié)小鼠脾臟T淋巴細(xì)胞亞群,提高IRF3表達(dá),促進(jìn)IL-10、IFN-γ的分泌,同時(shí)降低靶組織中TNF-α等炎癥因子。 結(jié)論：①甘露消毒丹是清代著名醫(yī)家葉天士所創(chuàng)之方,此方選藥精當(dāng),配伍嚴(yán)謹(jǐn),治療EV71感染濕熱證手足口病療效顯著,值得推廣。②甘露消毒丹能通過調(diào)節(jié)脾臟淋巴細(xì)胞亞群,顯著提高機(jī)體抗EV71病毒的免疫功能。③甘露消毒丹能誘導(dǎo)IRF3的表達(dá),刺激免疫細(xì)胞分泌IFN-γ干擾素提高機(jī)體的抗病毒能力,同時(shí)又能降低促炎因子TNF-α和提高抑炎IL-10細(xì)胞因子的分泌減輕炎癥反應(yīng)。④利殘方在調(diào)節(jié)免疫功能方面有優(yōu)勢(shì),清殘方在控制炎癥反應(yīng)方面更擅長,他們之間相輔相成。甘露消毒丹中清熱解毒藥、利濕化濁藥各有所長,各司其職,方劑配伍全面合理。
[Abstract]:Objective: to investigate the immunomodulatory effect of Ganlu Xiexian Dan (GXD) and its residual prescription on immune inflammation injury in mice infected with EV71, and to understand the scientific connotation of the compatibility of Ganlu Xiexian Dan (GXD). Methods: literature study: from the source of mannoxiao Dan, clinical application. Experimental study on the effectiveness of Gannuzhidan in the treatment of Hand-Foot-mouth Disease; ICR mice of 7 days of age were divided into blank group, model group, whole prescription group, Fang and remnant prescription group, and Qingruan recipe group. In the high temperature and high humidity environment, the EV71 virus model was made by intraperitoneal injection and the corresponding drug intervention was used. IL-10 and IFN- 緯 in the target tissues of mice were detected by Elisa method. Changes of cytokines such as TNF- 偽; The lymphocyte subsets of spleen were detected by flow cytometry. The expression of IRF3 protein in target tissues was detected by Western blot method. Results according to the literature study, Mannu disinfectant was created by Yetianshi. The clinical manifestation of mice infected with EV71 was observed on the 5th day after infection with EV71 type HFMD. The morphology and expression of EV71 virus in hind limb muscle were detected, which were in accordance with the model making standard. It was confirmed that the successful establishment of the model could regulate the spleen T lymphocyte subsets of mice with the successful establishment of 3. 3 mannose disinfectant and its residual prescription. Increase the expression of IRF3, promote the secretion of IL-10 and IFN- 緯, and decrease the inflammatory factors such as TNF- 偽 in target tissues. Conclusion 1 Mannu disinfectant Dan is the prescription of Ye Tianshi, a famous doctor in Qing Dynasty. The prescription is effective in treating hand-foot-mouth disease of EV71 infection with damp-heat syndrome. It is worth popularizing 2.Mannuxiaodan can significantly improve the immune function of the body against EV71 virus by regulating the lymphocyte subsets of spleen, and can induce the expression of IRF3. 3. Stimulating the immune cells to secrete IFN- 緯 interferon enhances the antiviral ability of the body. At the same time, it can reduce the inflammatory factor TNF- 偽 and increase the secretion of anti-inflammatory IL-10 cytokines. Qingruo prescription is better at controlling inflammatory reaction, and they complement each other. MannoxuoDan has its own strengths and advantages in heat clearing and detoxifying medicine, and it has its own advantages and responsibilities, and its compatibility is comprehensive and reasonable.
