正弦交變電磁場對體外培養(yǎng)大鼠股骨骨組織的影響

【摘要】:研究50Hz、1.8mT正弦交變電磁場(SEMFs)對體外培養(yǎng)大鼠股骨組織的影響。體外分離培養(yǎng)大鼠股骨組織,隨機(jī)分為兩組(磁場處理組和對照組)。磁場處理組用1.8mT的SEMFs每天處理股骨組織1.5h。在SEMFs處理后的第0d(第一次處理1.5h后)、1、2、3、4和5d,采用Real-time RT-PCR檢測培養(yǎng)股骨組織中骨形成相關(guān)基因表達(dá)量的變化;在SEMFs處理后的第3、6、9和12d測定培養(yǎng)股骨組織中堿性磷酸酶(ALP)活性變化;在SEMFs處理后的3、6、9和12d測定培養(yǎng)股骨中鈣含量的變化。結(jié)果表明,SEMFs能使OPG和Collagen-1mRNA表達(dá)量保持在一個(gè)相對穩(wěn)定的水平,在SEMFs處理后第1d和5d明顯增加Runx-2 mRNA的表達(dá)水平。在SEMFs處理后的3d和9dALP活性明顯增高(P0.01)。SEMFs處理培養(yǎng)股骨組織6、9和12d后,鈣含量明顯高于對照組(P0.01)。50Hz、1.8mT SEMFs能促進(jìn)體外培養(yǎng)大鼠股骨組織中骨代謝相關(guān)基因的表達(dá);增加鈣含量以及ALP活性。因此,本實(shí)驗(yàn)從體外培養(yǎng)股骨骨組織水平提供了SEMFs促進(jìn)骨代謝的實(shí)驗(yàn)依據(jù)。
[Abstract]:To study the effect of 50Hz and 1.8mT sinusoidal electromagnetic field (SEMFs) on the tissue of rat femur in vitro. The rat femur tissues were isolated and cultured in vitro, and were randomly divided into two groups (magnetic field treatment group and control group). The magnetic field treatment group treated 1.5h of the femur tissue with 1.8mT SEMFs every day. In the first 0d after SEMFs treatment (for the first time after 1.5h treatment), 1, 2, 3, 4 and 5D, using Real-time RT-PCR to detect the expression of genes related to culture the formation of femoral bone tissues; Determination of alkaline phosphatase in tissue culture of femur after treatment with SEMFs third, sixth, ninth and 12D (ALP) activity was measured; calcium content of femur cultured after SEMFs treatment in 3, 6, 9 and 12D. The results showed that SEMFs could maintain OPG and Collagen-1mRNA expression at a relatively stable level. After SEMFs treatment, 1D and 5D significantly increased the expression level of Runx-2 mRNA. The activity of 3D and 9dALP increased significantly after SEMFs treatment (P0.01). After SEMFs treatment of 6, 9 and 12D femur tissues, the content of calcium was significantly higher than that of the control group (P0.01). 50Hz, 1.8mT SEMFs can promote the expression of cultured rat bone bone metabolism related genes in vitro; increase the calcium content and the activity of ALP. Therefore, this experiment provides the experimental basis for SEMFs to promote bone metabolism from the level of bone tissue culture in vitro.
