Th17細(xì)胞及其細(xì)胞因子白細(xì)胞介素-17在粉塵螨致敏哮喘小鼠中的變化

【摘要】:目的探討Th17細(xì)胞及其細(xì)胞因子白細(xì)胞介素-17(IL-17)在粉塵螨致敏哮喘小鼠中的變化及其意義。方法 20只BALB/c小鼠隨機(jī)均分為哮喘組和健康對(duì)照組。哮喘組小鼠于第0、7和14天每鼠腹腔注射200μl致敏液[含粉塵螨粗浸液提取物50μg,Al(OH)32 mg]致敏。末次免疫后1周采用粉塵螨粗浸液提取物連續(xù)進(jìn)行滴鼻激發(fā),每天1次,每次50μg,激發(fā)7次,健康對(duì)照組給予等體積的PBS[含Al(OH)32 mg]處理。末次激發(fā)后24 h內(nèi)處死小鼠,取血清和肺泡灌洗液,無菌取脾臟。ELISA檢測(cè)血清中Ig G1、Ig E和肺泡灌洗液中IL-17的含量;流式細(xì)胞儀檢測(cè)脾臟中Th17細(xì)胞百分率。結(jié)果哮喘組小鼠血清中Ig G1和Ig E水平分別為(0.10±0.01)pg/ml和(1.15±0.10)pg/ml,高于健康對(duì)照組的(0.06±0.01)pg/ml和(0.04±0.01)pg/ml(P0.05);哮喘組小鼠肺泡灌洗液IL-17水平(85.13±2.36)pg/ml高于健康對(duì)照組(48.27±4.14)pg/ml(P0.01);哮喘組小鼠脾臟Th17細(xì)胞百分率(5.19±0.68)%高于健康對(duì)照組(0.95±0.19)%(P0.01),且脾臟Th17細(xì)胞百分率與肺泡灌洗液中IL-17水平呈正相關(guān)(r=0.851,P0.01)。結(jié)論粉塵螨致敏哮喘小鼠較健康小鼠肺泡灌洗液IL-17水平和脾臟Th17細(xì)胞數(shù)量均升高。
[Abstract]:Objective to investigate the changes and significance of Th17 cells and their cytokine IL-17 (IL-17) in asthma mice sensitized by Dermatophagoides farinae. Methods 20 BALB/c mice were randomly divided into asthma group and healthy control group. In the asthma group, 200 渭 l sensitized solution (50 渭 g, Al (OH) 32 mg) was injected intraperitoneally on the 0th, 7th and 14th day. One week after the last immunization, the crude extract of Dermatophagoides farinae was used for nasal stimulation once a day, 50 渭 g each time for 7 times. The healthy control group was treated with the same volume of PBS [containing Al (OH) 32 mg]. The mice were killed within 24 hours after the last challenge, serum and alveolar lavage fluid were taken, and spleen was taken aseptically. The contents of Ig G1, IgE and IL-17 in serum and alveolar lavage fluid were detected by Elisa, and the percentage of Th17 cells in spleen was detected by flow cytometry. Results the serum levels of Ig G 1 and Ig E in asthma group were (0.10 鹵0.001) pg/ml and (1.15 鹵0.10) pg/ml, respectively, which were higher than those in healthy control group (0.006 鹵0.01pg/ml and 0.004 鹵0.01pg/ml, P 0.05), and the levels of IL-17 in alveolar lavage fluid in asthma group (85.13 鹵2.36) pg/ml were higher than those in healthy control group (48.27 鹵4.14) pg/ml (P 0.01), and the levels of IL-17 in alveolar lavage fluid in asthma group were significantly higher than those in healthy control group (P < 0.01).
