間充質(zhì)干細(xì)胞在哮喘大鼠氣道炎癥及氣道重塑中的作用
[Abstract]:Objective to investigate the role of (MSCs) in chronic airway inflammation and airway remodeling in asthma rats. Methods 18 female SD rats were randomly divided into normal control group, asthma 4 week group and asthma 8 week group with 6 rats in each group. After ovalbumin (OVA) was sensitized, the asthma model was established by atomizing inhaled OVA. After the asthma model was successful, the airway pressure was measured. The (ESO) infiltration of eosinophils in rat airway smooth muscle was analyzed by HE staining and Image-Pro Plus image analysis software. The inner circumference, wall area, bronchial smooth muscle area and nucleus number of smooth muscle cells were measured. Peripheral blood mononuclear cells were isolated by density gradient centrifugation. The content of MSCs in each group was detected by flow cytometry and its relationship with airway inflammation and airway remodeling in asthma was analyzed. Results Airway responsiveness, airway wall EOS count, bronchial wall area, bronchial smooth muscle area, number of smooth muscle cells in asthma group and 8 weeks asthma group were studied. The proportion of MSCs in peripheral blood was significantly higher than that in the control group (P0.01, P0.05). There were significant differences in the above indexes between the two groups (P 0.01). Pearson linear correlation showed that the content of MSCs in peripheral blood mononuclear cells was positively correlated with airway responsiveness, EOS infiltration, bronchial wall area, bronchial smooth muscle thickness and bronchial smooth muscle cell nucleus (P01). Conclusion the increase of MSCs content in asthma may be involved in chronic airway inflammation and airway remodeling in asthma.
