低分子肝素、N-乙酰半胱氨酸干預(yù)對(duì)COPD大鼠氣道重塑的影響

【摘要】:目的探討低分子肝素(LMWH)、N-乙酰半胱氨酸(NAC)對(duì)慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)大鼠氣道重塑的影響。方法將40只6~8周鼠齡按雌雄各半的SD大鼠分為5組,即健康對(duì)照組(C組),熏香煙及脂多糖(LPS)方法制備的COPD模型組(M組),以及低分子肝素(LMWH)注射組(L組)和N-乙酰半胱氨酸(NAC)灌胃組(N組)及前二者同時(shí)干預(yù)組(LN組);光鏡下觀察大鼠病理學(xué)改變;以圖像分析系統(tǒng)測(cè)量其支氣管壁厚度;偏振光顯微鏡下觀察各組大鼠支氣管黏膜下膠原天狼猩紅染色的氣道膠原沉積。結(jié)果①模型制備:本研究制備的COPD模型大鼠支氣管、肺病理改變與人類COPD變化基本一致。②支氣管壁厚度:與對(duì)照組比較,M組、L組、N組和LN組管壁厚度明顯增加(P0.01);而與M組比較,L組、LN組管壁厚度均減少,且以LN組減少最為明顯(P0.01),N組亦有減少但差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。③膠原沉積:與對(duì)照組比較,各組主要呈現(xiàn)Ⅰ型膠原沉積,L組和N組膠原減少,LN組膠原減少更趨明顯。結(jié)論①較短時(shí)間的熏香煙+灌注LPS是建立COPD大鼠模型的簡(jiǎn)單易行的方法;②LMWH、NAC干預(yù)尤其是聯(lián)合應(yīng)用可通過不同作用機(jī)制減輕COPD發(fā)病過程中氣道壁厚度及氣道膠原沉積,進(jìn)而減輕COPD氣道重塑。
[Abstract]:Objective to investigate the effect of low molecular weight heparin (LMWH) (LMWH), N-acetylcysteine (NAC) on airway remodeling in rats with chronic obstructive pulmonary disease (chronic obstructive pulmonary disease,COPD). Methods 40 6-week-old SD rats were divided into 5 groups: healthy control group (C group), COPD model group prepared by smoking cigarette and lipopolysaccharide (LPS) method (M group), and divided into 5 groups: control group (C group) and COPD model group (M group) prepared by cigarette smoking and lipopolysaccharide (LPS) method. Low molecular weight heparin (LMWH) injection group (L group) and N-acetylcysteine (NAC) infusion group (N group) and the first two intervention groups (LN group); The pathological changes of rats were observed under light microscope, the thickness of bronchus wall was measured by image analysis system, and the airway collagen deposition by Sirius red staining was observed under polarized light microscope. Results 1 Model preparation: the pathological changes of bronchi and lung of the COPD model rats in this study were basically consistent with the changes of human COPD. 2 airway wall thickness: compared with the control group, group M, group L, the pathological changes of the bronchi and lung were similar to those of the human lung. The thickness of wall in N group and LN group was significantly increased (P0.01).
