反復接種BCG對慢性哮喘小鼠IL-33表達的影響

【摘要】:目的:研究在慢性哮喘模型中,BCG對哮喘的保護是否通過IL-33介導,為研發(fā)哮喘的新治療方法提供理論基礎。 方法:通過建立慢性哮喘小鼠模型,觀察三組(正常小鼠、慢性哮喘模型小鼠、BCG干預組)肺組織IL-33的mRNA表達,支氣管肺泡灌洗液及小鼠血清中IL-33表達。 結(jié)果:在慢性哮喘模型中,小鼠支氣管肺泡灌洗液(BALF)嗜酸性細胞計數(shù)在模型組(8.429±5.711*104/ml個)顯著高于對照組(0.286±0.488*104/ml個),說明慢性哮喘模型建模成功(n=7,P0.01)。實驗組(3.714±2.2887*104/ml個)顯著低于建模組(n=7,P0.05)。在慢性哮喘模型中,IL-33在模型組小鼠肺組織中的表達高于正常對照小鼠組4.154±1.020倍(n=6,P0.01)。實驗組小鼠肺組織中IL-33mRNA為對照組的2.791±1.230倍(n=6,P0.05) 結(jié)論:在慢性哮喘模型小鼠中,BCG干預可以降低肺部嗜酸性粒細胞浸潤。慢性哮喘模型小鼠肺組織IL-33mRNA顯著高于生理鹽水組,但BCG干預組的IL-33則顯著降低。提示BCG對哮喘的保護機制可能通過IL-33介導。
[Abstract]:Aim: to study whether the protection of BCG against asthma in chronic asthma model is mediated by IL-33, and to provide a theoretical basis for the development of a new treatment for asthma. Methods: the mRNA expression of IL-33, the expression of IL-33 in bronchoalveolar lavage fluid (BALF) and serum of three groups (normal mice, chronic asthmatic mice, BCG intervention group) were observed. Results: in the model of chronic asthma, the eosinophil count of (BALF) in the bronchoalveolar lavage fluid was significantly higher in the model group (8.429 鹵5.711*104/ml) than in the control group (0.286 鹵0.488*104/ml). The results showed that the model of chronic asthma was successfully established (n = 7, P 0.01). The experimental group (3.714 鹵2.2887*104/ml) was significantly lower than that of the model group (P 0.05). In chronic asthma model, the expression of IL-33 in lung tissue of model group was 4.154 鹵1.020 times higher than that of normal control group. IL-33mRNA in the lung tissue of the experimental group was 2.791 鹵1.230 times of that of the control group (P 0.05). Conclusion: in the model mice with chronic asthma, BCG intervention can reduce the infiltration of eosinophils in the lung. IL-33mRNA in lung tissue of chronic asthmatic mice was significantly higher than that of saline group, but IL-33 in BCG intervention group was significantly lower. The results suggest that the protective mechanism of BCG against asthma may be mediated by IL-33.
