致敏原劑量對(duì)哮喘模型免疫細(xì)胞活化及肺組織損傷的影響

【摘要】:目的研究致敏原的劑量對(duì)哮喘模型免疫細(xì)胞活化及肺組織損傷的影響。方法以卵蛋白為致敏原,采用腹腔注射[高劑量組1 mg/(只·次),低劑量組0.1 mg/(只·次)]聯(lián)合霧化吸入建立小鼠哮喘模型。ELISA分析血液中過敏相關(guān)免疫球蛋白IgE及細(xì)胞因子IL-4與IFN-γ的水平,免疫熒光及流式細(xì)胞術(shù)分析脾臟免疫細(xì)胞(包括T、B和DC細(xì)胞)的活化程度,HE染色分析肺組織的損傷情況。結(jié)果模型組小鼠血液中IgE及IL-4的水平明顯高于對(duì)照組(P0.05),且高劑量組高于低劑量組(P0.05)。但I(xiàn)FN-γ的水平模型組均低于對(duì)照組(P0.05),高、低劑量組間無差異(P0.05)。模型組小鼠脾臟細(xì)胞膜型CD28、CD86、CD80、CD40和MHC-Ⅱ的表達(dá)均高于對(duì)照組(P0.05),且高劑量組高于低劑量組(P0.05)。顯微鏡下觀察模型組小鼠肺組織出現(xiàn)炎性細(xì)胞浸潤(rùn)及支氣管壁增厚等病理改變,高劑量組更為嚴(yán)重。結(jié)論哮喘引發(fā)的免疫細(xì)胞活化和肺組織病理損傷與致敏原的劑量呈正相關(guān)。
[Abstract]:Objective to study the effects of the dose of allergen on the activation of immune cells and lung injury in asthmatic model. Methods ovalbumin was used as allergen and intraperitoneally injected [1 mg/] in high dose group. The asthmatic model was established in mice in low dose group (0. 1 mg/) combined with atomization inhalation. The levels of IgE and cytokines IL-4 and IFN- 緯 in blood were analyzed by ELISA. Immunofluorescence and flow cytometry were used to analyze the activation of splenic immune cells (including TGB and DC cells), and HE staining was used to analyze the injury of lung tissue. Results the levels of IgE and IL-4 in the blood of the model group were significantly higher than those of the control group (P0.05), and that of the high-dose group was higher than that of the low-dose group (P0.05).
