血清中TGF-β1、MDA、SOD和IL-12B、IL-6、IL-1β基因多態(tài)性與寧夏煤工塵肺的關(guān)系研究

【摘要】:目的研究白細(xì)胞介素-12B(1188A/C位點)、白細(xì)胞介素-6(634C/G位點)、白細(xì)胞介素-1β(511C/T位點)基因多態(tài)性與寧夏漢族煤工塵肺(CWP)的關(guān)系;測定塵肺患者血清中轉(zhuǎn)化生長因子-β1(TGF-β1)、丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性水平,以探討其在煤工塵肺發(fā)病中的作用和意義。 方法采用聚合酶鏈反應(yīng)-限制片段長度多態(tài)性(PCR-RFLP)法對120例煤工塵肺患者和120例接塵對照人群進行IL-12B基因1188A/C位點、IL-6基因634C/G位點和IL-1β基因511C/T位點多態(tài)性分型分析,并隨機抽取30例煤工塵肺患者和40例正常健康人群分別應(yīng)用硫代巴比妥酸(TBA)比色法、黃嘌呤氧化酶法和酶聯(lián)免疫吸附法(EL ISA)進行MDA含量、SOD活性和TGF-β1含量的測定。 結(jié)果基因IL-12B、IL-6、IL-1β多態(tài)位點的基因型在兩組對象中的分布符合Hardy一weinberg平衡,,工齡、工種分布和煙齡在煤工塵肺組與接塵對照組之間比較無統(tǒng)計學(xué)差異(均P>0.05)。IL-12B基因1188位點在煤工塵肺組和接塵對照組中檢出三種基因型AA、AC、CC型,這三種基因型在煤工塵肺組和接塵對照組間分布無顯著性差異(P>0.05);等位基因C在煤工塵肺組和接塵對照組中所占比例分別為45.8%和36.7%,差異有統(tǒng)計學(xué)意義(P<0.05);IL-1β (C-511T)位點的三種基因型在煤工塵肺組和接塵對照組間分布差異無統(tǒng)計學(xué)意義(P>0.05);等位基因在煤工塵肺組和和接塵對照組比較無統(tǒng)計學(xué)意義(P>0.05);煤工塵肺組IL-6基因634位點基因型CC和CG+GG分布頻率(66.7%、33.3%)和接塵對照組(52.5%、47.5%)比較差異有統(tǒng)計學(xué)意義(P㩳0.05);等位基因在兩組分布差異無統(tǒng)計學(xué)意義(P>0.05)。煤工塵肺患者血清中MDA、TGF-β1含量分別為(41.61士33.35)nmol/ml、(27.76士13.86) ng/ml,明顯高于健康人群(6.52士5.78)nmol/ml、(20.35士14.56)ng/ml,兩組比較差異均有有統(tǒng)計學(xué)意義(P0.05)。SOD活性煤工塵肺組(86.54士19.85) U/ml,明顯低于健康人群(104.87士18.08)U/ml,兩組比較差異有統(tǒng)計學(xué)意義(P0.05) 結(jié)論該研究發(fā)現(xiàn)IL-12B+1188A/C位點C等位基因可能是寧夏漢族煤工塵肺的易感基因之一。IL-6(-634C/G)位點多態(tài)性與寧夏地區(qū)煤工塵肺病的發(fā)生可能有關(guān)。IL-1β基因511位點多態(tài)性與寧夏地區(qū)煤工塵肺病的發(fā)生可能無關(guān)。寧夏煤工塵肺患者的MDA含量增高,SOD活力降低,細(xì)胞因子TGF-β1的表達增高,可能與煤工塵肺的發(fā)生發(fā)展有關(guān)。
[Abstract]:Objective to study the relationship between interleukin-12B (1188A/C), interleukin-6 (634C/G), interleukin-1 尾 (511C/T) gene polymorphisms and (CWP) in coal workers' pneumoconiosis in Ningxia Han nationality. The serum levels of transforming growth factor- 尾 1 (TGF- 尾 1), malondialdehyde (MDA) (MDA) and superoxide dismutase (SOD) in patients with pneumoconiosis were determined to explore the role and significance of transforming growth factor 尾 1 (TGF- 尾 1) and superoxide dismutase (SOD) in the pathogenesis of coal worker pneumoconiosis. Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to analyze the polymorphism of IL-12B gene 1188A/C locus, IL-6 634C/G locus and IL-1 尾 511C/T locus in 120 coal worker pneumoconiosis patients and 120 dust exposed controls. The activity of MDA and the content of TGF- 尾 1 were determined by thiobarbituric acid (TBA) colorimetry, xanthine oxidase assay and enzyme linked immunosorbent assay (EL ISA) in 30 coal worker pneumoconiosis patients and 40 normal controls. Results the genotypes of IL-12B,IL-6,IL-1 尾 polymorphic loci in the two groups were in accordance with the Hardy-weinberg equilibrium and the length of service. There was no significant difference in job type distribution and smoke age between the coal worker pneumoconiosis group and the dust exposed control group (P > 0. 05). Three genotypes of AA,AC,CC were detected in the coal worker pneumoconiosis group and the dust exposure control group at locus 1188 of IL-12B gene. There was no significant difference in the distribution of these three genotypes between the coal worker pneumoconiosis group and the dust-exposed control group (P > 0.05), and the proportion of allele C in the coal worker pneumoconiosis group and the dust exposure control group was 45.8% and 36.7%, respectively (P < 0.05). There was no significant difference in the distribution of IL-1 尾 (C-511T) genotypes between the coal worker pneumoconiosis group and the dust exposed control group (P > 0. 05), but there was no significant difference between the coal worker pneumoconiosis group and the dust exposure control group (P > 0. 05). The distribution frequency of CC and CG GG at 634 locus of IL-6 gene in coal worker pneumoconiosis group (66.7%) was significantly higher than that in dust-exposed control group (52.5% 47.5%). There was no significant difference in the distribution of alleles between the two groups (P > 0.05).
