不同劑量紅景天苷對COPD大鼠的骨骼肌功能及其FOXO1表達(dá)的影響

【摘要】:目的:探索不同劑量紅景天苷對COPD大鼠骨骼肌功能及其FOXO1表達(dá)的影響,為紅景天苷應(yīng)用于改善COPD骨骼肌功能障礙提供實(shí)驗(yàn)依據(jù)。方法:取40只成年SD大鼠分成5組,每組8只,分別為:空白組(C),COPD模型+安慰劑組(CS)、COPD模型+低劑量紅景天苷組(CS+L)、COPD模型+中劑量紅景天苷組(CS+M)、COPD模型+高劑量紅景天苷組(CS+H)。首先COPD模型+安慰劑組(CS)、COPD模型+低劑量紅景天苷組(CS+L)、COPD模型+中劑量紅景天苷組(CS+M)、COPD模型+高劑量紅景天苷組(CS+H)均采取單純被動吸煙發(fā)建立吸煙大鼠模型,空白組采取偽暴露法建立空白組模型?瞻捉M及COPD模型+安慰劑組(CS)給予安慰劑(生理鹽水)灌胃1個月,COPD模型+低劑量紅景天苷組(CS+L)、COPD模型+中劑量紅景天苷組(CS+M)、COPD模型+高劑量紅景天苷組(CS+H)分別予以低劑量(5mg/100g)、中劑量(25mg/100g)、高劑量(125mg/100g)紅景天苷灌胃處理1個月。記錄其體重變化,采用竭力游泳實(shí)驗(yàn)評估運(yùn)動能力,采集血清、肺組織、股四頭肌,對骨骼肌進(jìn)行稱重,測定骨骼肌樣本I型纖維比例,FOXO1、P-FOXO1、Mu RF-1、Atrogin-1in-1表達(dá)水平。結(jié)果:(1)體重比較:CS組(404.6±31.7g)較C組(475.6±20.4 g)下降,差異有統(tǒng)計學(xué)意義(P0.05);CS+L組(419.3±20.9 g)、CS+M組(425.6±32.6g)、CS+H組(432.7±27.2g)與CS組比較,體重均有所增加,并且各組值間值差異均有統(tǒng)計學(xué)意義(P0.05)。(2)竭力游泳時間比較:CS組(52.4±4.5min)較C組(14.9±4.1min)縮短,差異有統(tǒng)計學(xué)意義(P0.05);CS+L組(22.5±2.9min)、CS+M組(23.1±2.9min)、CS+H組(31.1±4.7min)與CS組比較,竭力游泳時間均有延長,并且各組值間值差異均有統(tǒng)計學(xué)意義(P0.05)(3)股四頭肌重量比較:CS組(2.82±0.22g)較C組(2.82±0.22g)減少,差異有統(tǒng)計學(xué)意義(P0.05);CS+L組(2.99±0.20g)、CS+M組(3.13±0.23g)、CS+H組(3.44±0.23g)與CS組比較,股四頭肌重量增加,并且各組值間值差異均有統(tǒng)計學(xué)意義(P0.05)(4)I型肌纖維比例比較:CS組(16.3±4.9%)較C組(32.8±2.8%)減少,差異有統(tǒng)計學(xué)意義(P0.05);CS+L組(22.2±3.8%)、CS+M組(25.7±3.9%)、CS+H組(28.2±2.9%)與CS組比較,I型肌纖維比例增加,并且各組值間值差異均有統(tǒng)計學(xué)意義(P0.05)(5)蛋白表達(dá)比較:(1)FOXO1:C組(0.260±0.10)、CS組(0.646±0.11)、CS+L組(0.440±0.13)、CS+M組(0.380±0.12)、CS+H組(0.330±0.01);(2)P-FOXO1:C組(0.230±0.06)、CS組(0.343±0.08)、CS+L組(0.401±0.06)、CS+M組(0.314±0.07)、CS+H組(0.289±0.06);(3)Mu RF-1:C組(0.338±0.077)、CS組(0.634±0.101)、CS+L組(0.428±0.067)、CS+M組(0.530±0.065)、CS+H組(0.353±0.105);(4)Atrogin-1:C組(0.393±0.013)、CS組(0.370±0.071)、CS+L組(0.306±0.103)、CS+M組(0.286±0.005)、CS+H組(0.157±0.061)。與CS組比較,FOXO1表達(dá)有所增加,并且各組值間值差異均有統(tǒng)計學(xué)意義(P0.05)。CS組與C組比較FOXO1、FOXO1、P-FOXO、Mu RF-1、的表達(dá)水平升高,差異均有統(tǒng)計學(xué)意義(P0.05);CS+L組、CS+M組、CS+H組與CS組比較,FOXO1、FOXO1、P-FOXO、Mu RF-1、Atrogin-1的表達(dá)水平下降,且影響水平與干預(yù)劑量呈正相關(guān),差異均有統(tǒng)計學(xué)意義(P0.05)。結(jié)論:1.紅景天苷可能通過降低FOXO1的表達(dá)保護(hù)COPD模型大鼠的骨骼肌功能,且這種保護(hù)作用為劑量依賴型。
[Abstract]:Objective: To explore the effects of different doses of salidroside on skeletal muscle function and FOXO1 expression in COPD rats, and to provide experimental evidence for salidroside application in improving skeletal muscle dysfunction of COPD. Methods: 40 adult SD rats were divided into 5 groups, 8 rats in each group: blank group (C), COPD model + placebo group (CS), COPD model + low dose of salidroside. First, COPD model + placebo group (CS), COPD model + low dose salidroside group (CS + L), COPD model + middle dose salidroside group (CS + M), COPD model + high dose salidroside group (CS + H) were used to establish smoking by passive smoking alone. Rats were given placebo (normal saline) for 1 month, COPD model + low dose salidroside group (CS + L), COPD model + middle dose salidroside group (CS + M), COPD model + high dose salidroside group (CS + H) for 5 mg / 100 g respectively. The body weight changes were recorded, the exercise ability was evaluated by exhaustive swimming test, the serum, lung tissue, quadriceps femoris were collected, the skeletal muscle was weighed, and the ratio of type I fibers, FOXO1, P-FOXO1, Mu RF-1, Atrogin-1 expression levels were measured in skeletal muscle samples. Weight comparison: CS group (404.6 + 31.7g) was significantly lower than C group (475.6 + 20.4g), the difference was statistically significant (P 0.05); CS + L group (419.3 + 20.9g), CS + M group (425.6 + 32.6g), CS + H group (432.7 + 27.2g) and CS group (432.7 + 27.2g) compared with CS group, the weight increased, and the difference between the values of each group was statistically significant (P 0.05). The weight of quadriceps femoris increased in CS + L group (2.99 + 0.20g), CS + M group (3.13 + 0.23g), CS + H group (3.44 + 0.23g) and CS group (3.44 + 0.23g), and the difference between the values of each group was statistically significant (P 0.05) (4) The proportion of type I muscle fibers in CS group (16.3 + 4.9%) was lower than that in C group (32.8 + 2.8%) (P 0.05). The percentage of type I myofibrils was increased in CS + H group (28.2 2 (2.9%) and CS + H group (28.2 (2.9%).
