急性肺損傷大鼠肺神經(jīng)導(dǎo)向因子Slit2及Robo4表達(dá)的研究

【摘要】:目的:觀察盲腸結(jié)扎穿孔致急性肺損傷(ALI)大鼠肺臟神經(jīng)導(dǎo)向因子Slit2及Robo4的改變,探討二者在ALI發(fā)生、發(fā)展中的作用。 方法:72只SD雄性大鼠隨機(jī)分為對(duì)照組(n=24)與ALI(n=48)組。ALI組采用盲腸結(jié)扎穿孔術(shù)復(fù)制ALI模型,對(duì)照組只開(kāi)腹、關(guān)腹,不行盲腸結(jié)扎穿孔術(shù)。于致傷后12、24、48h取大鼠動(dòng)脈血測(cè)定動(dòng)脈血氧分壓(PaO2);取肺測(cè)定濕/干質(zhì)量(W/D)比值、肺毛細(xì)血管通透性及組織病理學(xué)改變;酶聯(lián)免疫吸附法(ELISA)檢測(cè)血清腫瘤壞死因子(TNF-α)及白介素-6(IL-6)水平;逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)測(cè)定肺組織Slit2、Robo4mRNA表達(dá);免疫組織化學(xué)法觀察Slit2、Robo4蛋白表達(dá)情況。對(duì)照組每個(gè)時(shí)間點(diǎn)8只大鼠,ALI組每個(gè)時(shí)間點(diǎn)16只大鼠。 結(jié)果:與對(duì)照組比較,ALI組致傷后各時(shí)間點(diǎn)PaO2均顯著降低,肺W/D比值、肺毛細(xì)血管通透性、血清TNF-α及IL-6水平均顯著升高(P<0.05);病理觀察顯示肺組織受損;ALI組Slit2表達(dá)下降(P<0.05),Robo4表達(dá)差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。 結(jié)論:正常大鼠肺組織可表達(dá)Slit2及Robo4,,ALI大鼠肺組織Slit2表達(dá)下降,這可能與ALI發(fā)病有關(guān)。
[Abstract]:Aim: to observe the changes of Slit2 and Robo4 in the lung of acute lung injury (ALI) rats caused by cecal ligation and perforation, and to explore the role of them in the pathogenesis and development of ALI. Methods Seventy-two male SD rats were randomly divided into two groups: control group (n = 24) and ALI group (n = 48). The model of ALI was established by cecal ligation and perforation in Ali group. Arterial blood oxygen partial pressure (PaO2), wet / dry mass ratio (W / D), pulmonary capillary permeability and histopathological changes were measured at 12: 24 hours after injury. Serum levels of tumor necrosis factor (TNF- 偽) and interleukin-6 (IL-6) were detected by enzyme-linked immunosorbent assay (ELISA), the expression of Slit2,Robo4mRNA in lung tissue was detected by reverse transcription-polymerase chain reaction (RT-PCR), and the expression of Slit2,Robo4 protein was observed by immunohistochemistry. Control group (n = 8) and Ali group (n = 16). Results: compared with control group, PaO2 was significantly decreased, lung W / D ratio, pulmonary capillary permeability, serum TNF- 偽 and IL-6 levels were significantly increased in Ali group (P < 0. 05). There was no significant difference in Slit2 expression in ALI group (P < 0. 05). Conclusion: the expression of Slit2 and Slit2 in lung tissue of normal rats and Robo4,ALI rats can be decreased, which may be related to the pathogenesis of ALI.
