【摘要】：目的經(jīng)腹腔和氣管復(fù)制大鼠芥子氣(SM)急性肺損傷動(dòng)物模型,比較2種大鼠急性肺損傷模型肺纖維化指標(biāo)的差異。方法選取Sprague Dawley大鼠136只,隨機(jī)分為5組。腹腔SM組腹腔內(nèi)注入稀釋的SM 0.1 ml(0.96 LD50=8 mg/kg),氣管SM組氣管內(nèi)注入稀釋的SM 0.1 ml(0.98 LD50=2 mg/kg),腹腔和氣管丙二醇組分別腹腔和氣管內(nèi)注入丙二醇0.1 ml,正常對照組不做任何處理。采用Masson染色和免疫組織化學(xué),判斷肺纖維化指標(biāo)變化。結(jié)果 1光鏡下見72 h肺泡間隔被染成綠色的膠原纖維增多;2腹腔SM組大鼠肺泡間隔各時(shí)間段基質(zhì)金屬蛋白酶MMP-2、MMP-9、基質(zhì)金屬蛋白酶組織抑制劑TIMP-1、TIMP-2蛋白陽性表達(dá)率分別與氣管SM組比較升高(P0.05);3腹腔SM組大鼠肺泡間隔各時(shí)間段肺泡間隔Ⅰ型膠原、Ⅲ型膠原蛋白陽性表達(dá)率分別與氣管SM組比較升高(P0.05);4腹腔SM組大鼠肺泡間隔各時(shí)間段肺泡間隔轉(zhuǎn)化生長因子(TGF-β1)、母親DPP同源物7(Smad7)蛋白陽性表達(dá)率分別與氣管SM組比較升高(P0.05)。結(jié)論大鼠在經(jīng)腹腔和氣管SM LD50濃度相似的情況下,經(jīng)腹腔途徑肺纖維化指標(biāo)與經(jīng)氣管比較升高,提示SM經(jīng)腹腔途徑更容易誘導(dǎo)肺纖維化。
[Abstract]:Objective to establish a rat model of acute lung injury with mustard gas (SM) through abdominal cavity and trachea, and to compare the difference of pulmonary fibrosis indexes between two kinds of acute lung injury models. Methods 136 Sprague Dawley rats were randomly divided into 5 groups. The dilute SM 0.1 ml (0.96 LD50=8 mg/kg) was injected into the abdominal cavity of SM group, and the dilute SM 0.1 ml (0.98 LD50=2 mg/kg) was injected into the trachea in the SM group. The normal control group and the peritoneal and tracheal propanediol group were injected with 0.1 ml, in the abdominal cavity and trachea, respectively. The changes of pulmonary fibrosis were evaluated by Masson staining and immunohistochemistry. Results 1 under light microscope, the positive expression rate of matrix metalloproteinase (MMP-2,MMP-9,) MMP-2,MMP-9, tissue inhibitor of matrix metalloproteinase (MMP-2,MMP-9,) TIMP-1,TIMP-2 protein in the intraperitoneal SM group was observed at 72 h after the alveolar septum was stained green.
