高氧促進(jìn)不成熟肺組織HO-1表達(dá)的研究

【摘要】:目的研究持續(xù)高氧暴露時(shí),新生大鼠不成熟肺組織中血紅素加氧酶-1(HO-1)的表達(dá),推測(cè)其在高氧肺損傷機(jī)理中的作用。方法隨機(jī)將足月SD新生大鼠80只分為空氣組和高氧組,每組40只。高氧組出生后立即置入氧體積分?jǐn)?shù)95%的持續(xù)高氧環(huán)境中飼養(yǎng),空氣組則持續(xù)空氣中飼養(yǎng)。兩組分別于暴露于高氧或空氣中4、7、10d時(shí)隨機(jī)抽取8只,麻醉后取肺組織,比較兩組肺組織的病理學(xué)改變,采用免疫組化法檢測(cè)HO-1在肺組織中的表達(dá),并計(jì)算兩組肺組織面積積分光密度(IOD)值。結(jié)果病理學(xué)檢查顯示,高氧7d,可見(jiàn)肺泡結(jié)構(gòu)破壞,肺泡壁大量炎細(xì)胞侵潤(rùn),肺間隔增寬,可見(jiàn)明顯肺水腫改變。高氧10d,肺體積縮小,正常肺泡數(shù)目減少。肺泡結(jié)構(gòu)破壞,局部纖維增生,肺纖維化明顯。肺組織中血紅素加氧酶-1蛋白陽(yáng)性表達(dá)主要表現(xiàn)為肺泡上皮細(xì)胞、支氣管上皮細(xì)胞及肺泡巨噬細(xì)胞胞漿呈棕黃色,高氧4、7、10d組IOD值表明各時(shí)間點(diǎn)肺組織的HO-1表達(dá)均高于空氣組(P0.05)。結(jié)論持續(xù)高濃度氧可促進(jìn)不成熟肺組織HO-1表達(dá)增加,這種表達(dá)增加可能與不成熟肺組織高氧肺損傷病理生理有關(guān)。
[Abstract]:Objective to investigate the expression of heme oxygenase-1 (HO-1) in immature lung tissues of neonatal rats exposed to hyperoxia and to speculate its role in the mechanism of hyperoxia lung injury. Methods 80 adult Sprague-Dawley rats were randomly divided into air group and hyperoxia group with 40 rats in each group. The hyperoxia group was fed with 95% oxygen volume fraction immediately after birth, while the air group was kept in the air. Eight lung tissues were randomly selected after 10 days of exposure to hyperoxia or air. The pathological changes of lung tissue were compared between the two groups. The expression of HO-1 in lung tissue was detected by immunohistochemical method. The integral optical density (IOD) of lung tissue area in both groups was calculated. Results pathological examination showed that at 7 days of hyperoxia, the alveolar structure was destroyed, a large number of inflammatory cells in the alveolar wall invaded, the pulmonary septum widened, and obvious changes of pulmonary edema were observed. After 10 days of hyperoxia, the volume of lung decreased and the number of normal alveoli decreased. The alveolar structure was destroyed, local fibrous hyperplasia and pulmonary fibrosis were observed. The positive expression of heme oxygenase-1 protein in lung tissues was mainly found in alveolar epithelial cells, and the cytoplasm of bronchial epithelial cells and alveolar macrophages was brown. The IOD value of hyperoxia group for 10 days showed that the expression of HO-1 in lung tissue was higher than that in air group at each time point (P0.05). Conclusion continuous high oxygen concentration can promote the increase of HO-1 expression in immature lung tissues, which may be related to the pathophysiology of hyperoxia lung injury in immature lung tissues.
