紅霉素上調(diào)組蛋白去乙;2表達(dá)減輕氧化應(yīng)激下THP-1細(xì)胞的糖皮質(zhì)激素抵抗

【摘要】:目的探討紅霉素(EM)對(duì)氧化應(yīng)激下人THP-1單核細(xì)胞糖皮質(zhì)激素抵抗的作用及機(jī)制。方法 EM預(yù)孵育THP-1細(xì)胞后予煙草煙霧提取物(CSE)刺激細(xì)胞,用質(zhì)粒脂質(zhì)體法將構(gòu)建好的組蛋白去乙;2沉默RNA(HDAC2-siRNA)轉(zhuǎn)染細(xì)胞,ELISA檢測細(xì)胞培養(yǎng)上清白細(xì)胞介素8(IL-8)的含量,實(shí)時(shí)熒光定量PCR(qRT-PCR)和Western blot法分析HDAC2表達(dá)情況。結(jié)果 EM組的IL-8抑制率明顯高于CSE組,而低于對(duì)照組(P0.05),EM組地塞米松半數(shù)抑制濃度(IC50-Dex)低于CSE組,而高于對(duì)照組(P0.05)。EM組HDAC2蛋白的表達(dá)高于CSE組,而低于對(duì)照組(P0.05)。除此之外,應(yīng)用HDAC2-siRNA轉(zhuǎn)染細(xì)胞后HDAC2 mRNA及HDAC2蛋白的表達(dá)均明顯低于對(duì)照組及空轉(zhuǎn)組,而加用EM后其表達(dá)明顯升高(P0.05)。結(jié)論 CSE刺激下的THP-1細(xì)胞對(duì)地塞米松的敏感性明顯降低,EM可通過上調(diào)HDAC2蛋白的表達(dá)減輕氧化應(yīng)激下THP-1細(xì)胞的糖皮質(zhì)激素抵抗。
[Abstract]:Objective to investigate the effect and mechanism of erythromycin (EM) on glucocorticoid resistance of human THP-1 monocytes under oxidative stress. Methods THP-1 cells were preincubated with EM and stimulated by tobacco smoke extract (CSE). The constructed histone deacetylase 2 silencing RNA (HDAC2-siRNA) transfected cells were transfected with HDAC2-siRNA to detect the interleukin-8 (IL-8) content in the supernatant by plasmid liposome method. The expression of HDAC2 was analyzed by real time quantitative PCR (qRT-PCR) and Western blot. Results the inhibition rate of IL-8 in EM group was significantly higher than that in CSE group, but lower than that in control group (P0.05). IC50-Dex in EM group was lower than that in CSE group, while the expression of HDAC2 protein in EM group was higher than that in CSE group (P0.05), but lower than that in control group (P0.05). In addition, the expression of HDAC2 mRNA and HDAC2 protein in HDAC2-siRNA transfected cells were significantly lower than those in control group and empty group, but the expression of HDAC2 mRNA and HDAC2 protein were significantly increased after treatment with EM (P0.05). Conclusion the sensitivity of THP-1 cells stimulated by CSE to dexamethasone is significantly reduced by up-regulating the expression of HDAC2 protein to alleviate the glucocorticoid resistance of THP-1 cells under oxidative stress.
