氣道剪應力作用下黏蛋白5AC胞外極向分泌的主要介導機制研究

【摘要】:目的:探討氣道剪應力(SS)作用下黏蛋白(MUC)5AC胞外極向分泌的主要介導機制。 方法: (1)采用Cortactin-siRNA-pSilencer3.1-H1/neo表達載體轉染Cortactin-siRNA抑制Cortactin功能,并用Western印跡法檢測目標序列組與對照序列組的Cortactin水平,確認轉染成功后進行后續(xù)實驗。 (2)采用節(jié)律性旋轉裝置模擬呼吸時氣流介導的SS。該裝置旋轉過程中通過加速/減速交替進行的方式模擬呼吸過程中吸氣/呼氣對氣道的牽拉作用即為剪應力。為模擬病理狀態(tài)下氣流對氣道上皮細胞的SS作用,,我們把加速/減速的時間設置為1.2s/次,30次/min。每組作用30min。 (3)人支氣管上皮細胞(16HBE)以5×105個細胞/孔接種在6孔硅膠培養(yǎng)皿上并置于37℃、5%CO2培養(yǎng)箱中培養(yǎng),待細胞生長融合至70%左右,使用Stata軟件產(chǎn)生隨機數(shù)組隨機分為5組:A組(空白對照組);B組(SS組);C組(SS+Rac-1特異性抑制劑組);D組(SS+F-actin解聚劑組);E組(SS+Cortactin-siRNA轉染組);每組各設6個復孔。置于37℃、5%CO2培養(yǎng)箱中繼續(xù)培養(yǎng)2h后進行相關檢測。上述實驗重復3次用于統(tǒng)計學分析。 (4)分別采用酶聯(lián)免疫吸附(ELISA)法測定細胞培養(yǎng)上清液中MUC5AC的胞外相對分泌量; Western印跡法檢測Cortactin及p-Cortactin的相對水平和轉染效果;激光共聚焦顯微鏡觀察F-actin聚合情況。 結果: (1)通過Western印跡法檢測目標序列組與對照序列組的Cortactin相對表達水平后確認Cortactin-siRNA-pSilencer3.1-H1/neo表達載體轉染成功,可用于后續(xù)實驗。 (2) ELISA法檢測各組細胞上清液中MUC5AC水平:與A組(0.210±0.013)相比,B組(0.631±0.025)MUC5AC胞外相對分泌水平顯著增加(P=0.000);與B組相比,C組、D組和E組(0.473±0.112、0.330±0.067、0.272±0.019)MUC5AC胞外相對分泌水平均明顯降低(P=0.043、0.000、0.000)。 (3) Western印跡法檢測Cortactin及p-Cortactin的相對水平:B組Cortactin相對表達水平為(0.670±0.048),顯著高于E組的(0.132±0.014)(P<0.01),但與A、C、D組的(0.641±0.016、0.622±0.012、0.653±0.027)比較差異均無統(tǒng)計學意義(均P>0.05);B組p-Cortactin相對表達水平為(0.582±0.067),顯著高于A、C、E組的(0.131±0.011、0.393±0.045、0.170±0.016)(P=0.000、0.021、0.000),而D組(0.511±0.029)與B組差異無統(tǒng)計學意義(P=0.246)。 (4)激光共聚焦顯微鏡觀察F-actin聚合情況:A組幾乎沒有聚合狀態(tài)的F-actin,表現(xiàn)為無絲狀熒光出現(xiàn);B組F-actin聚合程度高,呈平行排列、粗細均勻、長短一致的絲狀熒光;C組F-actin聚合程度較B組略有減弱,絲狀熒光的平行排列稍顯紊亂、粗細稍欠均勻、長短不一致;D組F-actin聚合程度顯著降低,絲狀熒光排列紊亂,無細長的絲狀熒光;E組F-actin聚合程度明顯降低,絲狀熒光長短不一、粗細不均、排列紊亂。 結論: 在氣道SS作用下,通過激活Rac-1、Cortactin和F-actin而促進MUC5AC胞外極向分泌,產(chǎn)生黏液高分泌的病理狀態(tài)。Rac-1、Cortactin和F-actin是SS作用下MUC5AC胞外極向分泌的主要介導分子。
[Abstract]:Aim: to investigate the mediating mechanism of extracellular polar secretion of MUC5 AC under the action of airway shear stress (SSSS). Methods: 1) Cortactin-siRNA-pSilencer3.1-H1/neo expression vector was used to transfect Cortactin-siRNA to inhibit the function of Cortactin. The level of Cortactin in target sequence group and control sequence group was detected by Western blotting, and the subsequent experiment was carried out after the transfection was successful. A rhythmic rotating device was used to simulate the airflow mediated SSs during respiration. In the process of rotation, shear stress is simulated by alternate acceleration and deceleration in the process of breathing, in which the pulling effect of inspiratory and exhalation on the airway is simulated. In order to simulate the SS effect of airflow on airway epithelial cells in pathological state, we set the acceleration / deceleration time to 30 1.2s/ min.
