【摘要】：目的研究藻酸雙酯鈉(PSS)對脂多糖(LPS)所致的小鼠肺微血管內皮細胞(PMVEC)損傷的保護作用。方法體外培養(yǎng)PMVEC,隨機將細胞分為空白對照組(C組)、LPS刺激組(L組)、PSS+LPS組(LP組)。采用MTT法檢測PSS對LPS刺激PMVEC的細胞活力的影響,虎紅染色法測定中性粒細胞(PMN)對PMVEC黏附數(shù)量的影響,酶聯(lián)免疫吸附試驗(ELISA)法檢測3組培養(yǎng)上清液中腫瘤壞死因子-α(TNF-α)和細胞間黏附分子-1(ICAM-1)的濃度。結果 PSS能抑制LPS所致的PMVEC細胞活力下降(P=0.001);與C組比較,L組的PMVEC與PMN的黏附數(shù)量明顯增加(P=0.000),TNF-α和ICAM-1的表達顯著增加(P=0.000);與L組比較,PSS預處理1h能顯著降低LPS所致的PMVEC與PMN的黏附(P=0.000),LP組的TNF-α和ICAM-1表達顯著降低(P0.05)。結論 PSS能抑制LPS對PMVEC的損傷及PMVEC與PMN的黏附,其作用機制可能與降低ICAM-1和TNF-α的表達有關。
[Abstract]:Objective to study the protective effect of sodium alginate (PSS) on PMVECs induced by lipopolysaccharide (LPS) in mice pulmonary microvascular endothelial cells (PMVECs). Methods PMVECs were cultured in vitro. The cells were randomly divided into two groups: control group (C group), LPS-stimulated group (L group) and PSS LPS group (LP group). MTT assay was used to detect the effect of PSS on the cell viability of PMVEC stimulated by LPS, and the effect of neutrophil PMNs on the adhesion of PMVEC was determined by tiger red staining. The concentrations of tumor necrosis factor- 偽 (TNF- 偽) and intercellular adhesion molecule-1 (ICAM-1) in the supernatants of the three groups were detected by Elisa. Results PSS could inhibit the decrease of PMVEC cell viability induced by LPS, and the adhesion between PMVEC and PMN in group L was significantly increased compared with group C, and the expression of TNF- 偽 and ICAM-1 was significantly increased in group L, and the PMVEC induced by LPS was significantly decreased after 1 h of pretreatment with PSS compared with group L. The expression of TNF- 偽 and ICAM-1 in LP group was significantly lower than that in P0. 05 group. Conclusion PSS can inhibit the damage of LPS to PMVEC and the adhesion of PMVEC to PMN, and its mechanism may be related to the decrease of ICAM-1 and TNF- 偽 expression.
