【摘要】：目的觀察腫瘤壞死因子(TNF-α)對急性肺損傷動物模型外周血骨髓間充質(zhì)干細胞(MSCs)數(shù)量的影響。方法 SD大鼠分為對照組、模型組和干預組。對照組給予生理鹽水氣道滴入;模型組給予內(nèi)毒素(LP S 5 mg/kg)氣道內(nèi)滴入;干預組氣道內(nèi)滴入LPS同時英夫利昔(10 mg/mL)腹腔注射5 mg/kg。5 h后處死動物取外周血,檢測TNF-α含量及成纖維細胞集落形成單位(CFU-F)數(shù)。成骨、成脂誘導檢測其分化特性。結(jié)果模型組大鼠外周血TNF-α水平較對照組顯著升高(354.0±82.4 vs 38.0±21.6,P0.05),而較干預組顯著降低(206.5±64.1 vs 354.0±82.4,P0.05)。模型組大鼠外周血中培養(yǎng)出的CFU-Fs明顯高于對照組(3.60±1.47vs 1.40±2.83,P0.05)及干預組(3.60±1.47 vs 1.70±1.72,P0.05);外周血骨髓MSCs成骨、成脂誘導后染色均陽性。結(jié)論急性肺損傷時,外周血中增加的TNF-α含量對骨髓MSCs動員及歸巢均有促進作用。
[Abstract]:Objective to observe the effect of TNF- 偽 on the number of bone marrow mesenchymal stem cells (MSCs) in acute lung injury model. Methods SD rats were divided into control group, model group and intervention group. The control group was given normal saline, the model group was given endotoxin LP S 5 mg / kg, the intervention group was injected with LPS and Inflexixide 10 mg / mL intraperitoneally for 5 mg/kg.5, and the animals were killed to collect the peripheral blood after 5 mg/kg.5. The content of TNF- 偽 and the number of fibroblast colony forming units (CFU-F) were measured. Osteogenesis and adipogenic induction were used to detect its differentiation characteristics. Results compared with the control group, the level of TNF- 偽 in the model group was significantly higher than that in the control group (354.0 鹵82.4 vs 38.0 鹵21.6P0.05), while the level of TNF- 偽 was significantly lower than that in the intervention group (206.5 鹵64.1 vs 354.0 鹵82.4 P0.05). The CFU-Fs cultured in the peripheral blood of the model group was significantly higher than that of the control group (3.60 鹵1.47vs 1.40 鹵2.83 P0.05) and the intervention group (3.60 鹵1.47 vs 1.70 鹵1.72 P0.05), and the MSCs of peripheral blood was positive in osteogenesis and lipogenic induction. Conclusion during acute lung injury, the increased TNF- 偽 content in peripheral blood can promote bone marrow MSCs mobilization and homing.
