【摘要】：目的探討B(tài)細胞活化轉(zhuǎn)錄因子(BATF)、維甲酸相關(guān)核孤兒受體γt(RORγt)、白細胞介素-17(IL-17)在哮喘小鼠脾組織中的表達及意義。方法將15只小鼠隨機分為3組:生理鹽水(NS)組、哮喘(AS)組和地塞米松(DXM)組,每組5只。利用卵清蛋白(OVA)造模。收集支氣管肺泡灌洗液(BALF)行白細胞分類計數(shù);HE染色觀察肺組織病理改變;酶聯(lián)免疫吸附測定法(ELISA)檢測脾組織勻漿上清液IL-17的質(zhì)量濃度;實時熒光定量聚合酶鏈反應(yīng)(RT-PCR)檢測脾組織中BATF、RORγt mRNA的表達量;并對上述指標的相關(guān)性進行分析。結(jié)果 1)與AS組相比,DXM組小鼠BALF白細胞(WBC)總數(shù)、中性粒細胞(NEU)數(shù)和嗜酸性粒細胞(EOS)數(shù)明顯降低(P0.01)。2)DXM組肺部炎癥較AS組明顯減輕。3)與AS組相比,DXM組脾組織勻漿上清液IL-17的質(zhì)量濃度減低(P0.05)。4)與AS組相比,DXM組脾組織中BATF、RORγt mRNA表達量減少(P0.01)。5)脾組織勻漿上清液IL-17的質(zhì)量濃度與BALF中WBC總數(shù)、NEU數(shù)、EOS數(shù)呈正相關(guān)(r=0.838、0.737、0.882,P均0.005);脾組織BATF mRNA的表達量與RORγt mRNA表達量、脾組織勻漿上清液IL-17的質(zhì)量濃度呈正相關(guān)(r=0.807、0.833,P均0.005);脾組織RORγt mRNA的表達量與脾組織勻漿上清液IL-17的質(zhì)量濃度呈正相關(guān)(r=0.890,P0.005)。結(jié)論 1)在哮喘小鼠脾組織中BATF與RORγt可能共同調(diào)節(jié)IL-17的分泌,并通過IL-17的致炎作用參與哮喘氣道炎癥的發(fā)生發(fā)展。2)DXM可能通過BATF、RORγt途徑部分阻止脾源性Th17細胞的分化,減少IL-17的分泌。
[Abstract]:Objective to investigate the expression and significance of B cell activated transcription factor (BATFN), the orphan receptor 緯 t(ROR 緯 T of retinoic acid and interleukin-17 (IL-17) in the spleen of asthmatic mice. Methods Fifteen mice were randomly divided into three groups: NS group, ASA group and DXM group with 5 rats in each group. Ovalbumin (ovalbumin) was used to make the model. BALF was collected from bronchoalveolar lavage fluid (BALF) to observe the pathological changes of lung tissue by leucocyte classification and HE staining, and the IL-17 concentration in spleen tissue homogenate supernatant was detected by enzyme linked immunosorbent assay (Elisa). Real-time fluorescence quantitative polymerase chain reaction (RT-PCR) was used to detect the expression of BATFFU ROR 緯 t mRNA in splenic tissues, and the correlation of the above indexes was analyzed. Results 1) compared with as group, the total number of BALF WBC in DXM group was higher than that in as group. The number of neutrophils (neu) and eosinophilic granulocytes (EOS) were significantly reduced in the DXM group than in the as group. 3) the mass concentration of IL-17 in the splenic tissue homogenate supernatant of the DXM group was significantly lower than that in the as group. 4) the BATFROR 緯 t in the spleen tissue of the DXM group was significantly lower than that in the as group. The expression of mRNA in the supernatant of splenic tissue homogenate was positively correlated with the total number of WBC and the number of WBC in BALF (P 0.005), and the expression of BATF mRNA in splenic tissue was correlated with the expression of ROR 緯 t mRNA. There was a positive correlation between the mass concentration of IL-17 in the supernatant of spleen tissue and the expression of ROR 緯 t mRNA in spleen tissue and the mass concentration of IL-17 in splenic tissue homogenate supernatant. Conclusion 1) BATF and ROR 緯 t may co-regulate the secretion of IL-17 in the splenic tissues of asthmatic mice, and participate in the occurrence and development of asthma airway inflammation through the inflammatory effect of IL-17. 2 DXM may partially block the differentiation of splenic Th17 cells through the BATFU ROR 緯 t pathway. Reduce the secretion of IL-17.
