【摘要】：目的:觀察丙酮酸乙酯(EP)對(duì)甲苯二異氰酸酯(TDI)誘導(dǎo)的哮喘小鼠氣道上皮細(xì)胞黏附連接蛋白E-cadherin表達(dá)的影響。方法:BALB/c小鼠30只,隨機(jī)分為3組。哮喘組第1天、第8天予0.3%TDI耳背致敏,第15、18、21天予3%TDI激發(fā),每次激發(fā)前1 h按100 mg/kg腹腔注射生理鹽水。對(duì)照組第1天、第8天予AOO(丙酮和橄欖油)致敏,余同哮喘組。EP治療組,同哮喘組,但在激發(fā)前1 h按100 mg/kg腹腔注射EP。第22天檢測(cè)各組小鼠的氣道高反應(yīng)性、淋巴上清IL-4、IFN-γ及血清Ig E,用免疫組化及WB測(cè)定肺組織E-cadherin。結(jié)果:哮喘組氣道高反應(yīng)性、淋巴上清IL-4、IFN-γ及血清Ig E均高于對(duì)照組,EP干預(yù)后顯著降低。免疫組化及免疫印跡結(jié)果示,對(duì)照組E-cadherin均勻致密分布于氣道上皮細(xì)胞連接處,哮喘組Ecadherin分布混亂、減少,EP干預(yù)能顯著減少E-cadherin的破壞。結(jié)論:EP可以減少TDI誘導(dǎo)的哮喘小鼠氣道上皮細(xì)胞E-cadherin的破壞。
[Abstract]:Objective: To observe the effect of ethyl pyruvate (EP) on the expression of adhesion connexin E-cadherin in airway epithelial cells in asthmatic mice induced by toluene diisocyanate (TDI). Methods: 30 BALB/c mice were randomly divided into 3 groups. The asthma group was first days, eighth days to 0.3%TDI ear sensitization, 3%TDI excitation on the day 15,18,21, and 100 mg/kg celiac before each stimulation of 1 h. The control group was injected with AOO (acetone and olive oil) on the eighth day for first days. The control group was treated with asthma group.EP and asthma group, but the airway hyperresponsiveness of each group was detected by 100 mg/kg intraperitoneal injection of EP. twenty-second days before the stimulation of 1 h. Lymphatic supernatant IL-4, IFN- gamma and Ig E in blood, immunohistochemistry and WB were used to determine the E-cadherin. results of lung tissue. The airway hyperresponsiveness, IL-4, IFN- gamma and serum Ig E in the asthmatic group were all higher than those in the control group, and the results of EP intervention were significantly lower. The immunohistochemical and Western blot results showed that the E-cadherin in the control group was uniformly distributed at the junction of the airway epithelial cells, the Ecadherin distribution in the asthmatic group was chaotic and decreased, and the EP intervention could significantly reduce the disruption of E-cadherin. Conclusion: EP can reduce the destruction of E-cadherin in airway epithelial cells of TDI induced asthmatic mice.
