谷氨酰胺對(duì)克雷伯桿菌引起的大鼠肺炎炎癥的抑制作用

【摘要】:目的探討谷氨酰胺對(duì)克雷伯桿菌引起的大鼠肺炎炎癥的抑制作用及機(jī)制。方法將36只SD大鼠隨機(jī)分為3組,正常組、模型組、谷氨酰胺組,通過(guò)氣管滴入克雷伯桿菌建立大鼠肺炎模型,谷氨酰胺組在造模前1d即腹腔注射10mL/kg谷氨酰胺,正常組和模型組給予等量生理鹽水,連續(xù)給藥7d,大鼠脫頸處死,取肺組織及血清進(jìn)行檢測(cè)。稱量記錄右肺濕重及干重(W/D),HE染色檢測(cè)肺組織病理形態(tài),酶聯(lián)免疫吸附試驗(yàn)(ELISA)法檢測(cè)肺組織勻漿及血清中腫瘤壞死因子-α(TNF-α)、白細(xì)胞介素-1β(IL-1β)、白細(xì)胞介素-6(IL-6)含量,RT-PCR檢測(cè)核因子-κB(NF-κB)p65和NF-κB抑制蛋白(IκBα)mRNA表達(dá),Western blot檢測(cè)NF-κB p6和IκBα蛋白表達(dá)。結(jié)果與模型組比較,谷氨酰胺組能降低W/D比值[5.98±0.29 vs.4.32±0.33(P0.05)],減輕肺組織充血水腫、炎性細(xì)胞浸潤(rùn),改善肺組織形態(tài),抑制血清和肺組織勻漿中TNF-α、IL-1β及IL-6的分泌(P0.05),并抑制NF-κB p65和IκBα磷酸化水平(P0.05)。結(jié)論谷氨酰胺能抑制克雷伯桿菌引起的大鼠肺炎炎癥,與抑制NF-κB信號(hào)通路有關(guān)。
[Abstract]:Objective to investigate the inhibitory effect and mechanism of glutamine on pneumonia induced by Klebsiella in rats.Methods Thirty-six SD rats were randomly divided into 3 groups: normal group, model group and glutamine group. The model of pneumonia was established by instillation of Klebsiella in trachea. 10mL/kg glutamine was injected intraperitoneally in the glutamine group 1 day before the model was made.The normal group and the model group were given the same amount of normal saline for 7 days. The rats were killed without neck, and the lung tissue and serum were detected.The right lung wet weight and dry weight were measured by HE staining to detect lung histopathology.The expression of TNF- 偽 TNF- 偽, IL-1- 尾 -IL-1 尾 and IL-6IL-6 in lung homogenate and serum were detected by Elisa. The expression of NF-魏 B-NF- 魏 B)p65 and NF- 魏 B inhibitory protein I 魏 B 偽 mRNA was detected by RT-PCR. The expression of NF- 魏 B p6 and I 魏 B 偽 was detected by Western blot.Results compared with the model group, the glutamine group could reduce the W / D ratio [5.98 鹵0.29 vs.4.32 鹵0.33 vs.4.32 鹵0.33 P0.05], alleviate the hyperemia and edema of lung tissue, inflammatory cell infiltration, and improve the morphology of lung tissue.It inhibited the secretion of TNF- 偽 IL-1 尾 and IL-6 in serum and lung tissue homogenate, and inhibited the phosphorylation levels of NF- 魏 B p65 and I 魏 B 偽.Conclusion Glutamine can inhibit the inflammation of pneumonia induced by Klebsiella in rats, which is related to the inhibition of NF- 魏 B signaling pathway.
