慢性阻塞性肺疾病患者外周血單個核細胞絲裂原活化蛋白激酶p38的表達

【摘要】:目的研究絲裂原活化蛋白激酶p38(p38 MAPK)在慢性阻塞性肺疾病(COPD)患者外周血單個核細胞中的表達情況及其與氣流受限的相關性。方法選擇確診的急性加重期COPD(AECOPD)患者50例作為實驗組,根據肺功能檢測,分為重-極重度組(A組)26例和輕-中度組(B組)24例。選擇同期門診健康受試者25例為對照組,采集各組受試者外周血,運用PCR熒光檢測法測定P38 MAPK mRNA的表達,酶聯(lián)免疫吸附法(ELISA)檢測外周血白細胞介素(IL)-6和IL-8含量。結果實驗組患者外周血單個核細胞核內p38 MAPK mRNA表達和血清IL-6、IL-8含量均高于對照組(P0.05);A組患者外周血單個核細胞核內p38 MAPK mRNA的表達和血清IL-6、IL-8含量顯著高于B組(P0.05);AECOPD患者外周血單個核細胞核內p38 MAPK mRNA表達隨1 s用力呼氣容積(FEV1)占預計值升高而下降(r=-0.91,P0.05)。結論 p38 MAPK、IL-6、IL-8參與了COPD的炎癥反應,且同COPD患者的氣流受限程度呈正相關。
[Abstract]:Objective to investigate the expression of mitogen-activated protein kinase (p38(p38) in peripheral blood mononuclear cells (PBMC) of patients with chronic obstructive pulmonary disease (COPD) and its correlation with airflow limitation.Methods 50 patients with acute exacerbation were selected as experimental group. According to the pulmonary function test, they were divided into two groups: group A (n = 26) and group B (n = 24).In the same period, 25 healthy subjects were selected as control group. Peripheral blood samples were collected from each group. The expression of P38 MAPK mRNA was detected by PCR fluorescence assay, and the levels of IL-6 and IL-8 in peripheral blood were detected by enzyme linked immunosorbent assay (Elisa).Results the expression of p38 MAPK mRNA in peripheral blood mononuclear nuclei and the level of serum IL-6 IL-8 in peripheral blood mononuclear nuclei of patients in the experimental group were higher than those in the control group P0.05AECOPD patients. The expression of p38 MAPK mRNA in the peripheral blood mononuclear nuclei and the level of serum IL-6 IL-8 in the peripheral blood mononuclear nuclei of patients with AECOPD were significantly higher than those in the patients with P0.05AECOPD.The expression of p38 MAPK mRNA in peripheral blood mononuclear cells decreased with the increase of forced expiratory volume (FEV1).Conclusion p38 MAPK- IL-6 IL-8 is involved in the inflammatory reaction of COPD, and is positively correlated with the degree of airflow limitation in COPD patients.
