富硒酵母對(duì)大氣細(xì)顆粒物致大鼠肺損傷的干預(yù)作用

【摘要】:目的探討富硒酵母對(duì)大氣細(xì)顆粒物(PM_(2.5))致大鼠肺損傷的干預(yù)作用。方法 56只健康成年SD大鼠隨機(jī)平均分為7組:生理鹽水對(duì)照組(氣管滴注生理鹽水);PM_(2.5)染毒組(氣管滴注PM_(2.5)混懸液,40 mg/kg,隔天一次,共3次);溶劑對(duì)照組(1%羧甲基纖維素灌胃);低、中、高劑量干預(yù)組(氣管滴注PM_(2.5)混懸液,末次滴注24h后,分別灌胃給予富硒酵母8.75、17.5和35 mg/kg,每天1次,共7d);干預(yù)對(duì)照組(35 mg/kg富硒酵母灌胃)。末次灌胃24 h后,收集肺泡灌洗液(BALF),測(cè)定BALF中腫瘤壞死因子-α(TNF-α)、白細(xì)胞介素-1β(IL-1β)、丙二醛(MDA)、總蛋白(TP)含量及總超氧化物歧化酶(T-SOD)、谷胱甘肽過(guò)氧化物酶(GSH-Px)、乳酸脫氫酶(LDH)及堿性磷酸酶(AKP)活性,并計(jì)數(shù)細(xì)胞。未灌洗肺葉制作病理切片。結(jié)果與對(duì)照組相比,PM_(2.5)染毒組及低、中、高劑量干預(yù)組BALF中中性粒細(xì)胞百分比、TNF-α、IL-1β、TP及MDA含量,LDH及AKP活性增高,GSH-Px及T-SOD活性下降(P0.05)。與PM_(2.5)染毒組相比,干預(yù)組BALF中中性粒細(xì)胞百分比、TNF-α、IL-1β、TP及MDA含量,LDH及AKP活性降低,GSH-Px及T-SOD活性升高(P"0.05),隨干預(yù)劑量增加,中性粒細(xì)胞百分比、TNF-α、IL-1β、TP及MDA含量,LDH及AKP活性降低,GSH-Px、T-SOD活性升高幅度增加。結(jié)論富硒酵母可適當(dāng)減輕PM_(2.5)所致大鼠肺組織炎性損傷及氧化應(yīng)激損傷。
[Abstract]:Objective to investigate the effect of selenium enriched yeast on lung injury induced by PMN 2.5 in rats.Methods 56 healthy adult Sprague-Dawley rats were randomly divided into 7 groups: saline control group (intratracheal drip of normal saline and PM2. 5) treated group (tracheal drip of PM2. 5) suspension 40 mg / kg, once every other day.The control group was fed with Se-enriched yeast for 35 mg/kg.24 hours after the last gavage,The contents of tumor necrosis factor- 偽 (TNF- 偽), interleukin-1 尾 (IL-1 尾), malondialdehyde (malondialdehyde) (MDAA), total protein (TTP) and the activities of total superoxide dismutase (T-SOD), glutathione peroxidase (GSH-PX), lactate dehydrogenase (LDH) and alkaline phosphatase (AKP) in BALF were measured and counted.The lung lobes were not lavaged to make pathological sections.The percentage of neutrophils in BALF in the intervention group decreased the activities of GSH-Px and T-SOD, and the contents of MDA, TP and MDA decreased, and increased with the increase of the dose of TNF- 偽, IL-1 尾, TNF- 偽, TNF- 偽, IL-1 尾, TNF- 偽, and the activity of T-SOD, and the activity of GSH-Px and T-SOD increased with the increase of the intervention dose.Percentage of neutrophilic granulocytes, TNF- 偽 -IL-1 尾, TP, and MDA content. LDH and AKP activities decreased and the activity of GSH-PxX T-SOD increased.Conclusion Selenium-enriched yeast can reduce the inflammatory injury of lung tissue and oxidative stress injury induced by PMSt 2.5 in rats.
