【摘要】：目的：明確慢性阻塞性肺疾病(COPD)大鼠股四頭肌中各型纖維的比例,及腫瘤壞死因子樣凋亡微弱誘導(dǎo)劑(TWEAK)、過氧化物酶體增生激活受體Y協(xié)同刺激因子-1α(PGC-1α)在COPD大鼠股四頭肌中的表達(dá)。初步探討是否由于COPD大鼠股四頭肌中TWEAK表達(dá)升高,導(dǎo)致PGC-1α表達(dá)降低,進(jìn)一步引起股四頭肌纖維類型發(fā)生轉(zhuǎn)換。 方法：將50只Wistar大鼠隨機(jī)分為對(duì)照組20只、COPD模型組30只；COPD模型組大鼠采用煙熏+氣管內(nèi)滴注豬胰彈性蛋白酶方法,共三個(gè)月；HE染色觀察肺和氣道組織病理變化、肺功能及大鼠一般狀況判斷造模是否成功。取大鼠股四頭肌稱重,制成冰凍切片,采用肌球蛋白ATP酶染色法,圖像分析儀計(jì)數(shù),觀察測(cè)量骨骼肌肌纖維Ⅰ型和Ⅱ型比例；免疫組化法及Western blot法分別測(cè)定股四頭肌TWEAK、PGC-1α蛋白表達(dá)水平。所得數(shù)據(jù)經(jīng)SPSS17.0軟件做正態(tài)性檢驗(yàn)、t檢驗(yàn)以及相關(guān)性分析。 結(jié)果： 1.通過進(jìn)行大鼠肺功能測(cè)量,以及觀察肺組織的病理切片,判定COPD模型組大鼠肺符合COPD診斷標(biāo)準(zhǔn)。 2.COPD模型組大鼠體重以及股四頭肌的重量較正常對(duì)照組均有明顯減輕(P0.05)。 3.COPD模型組大鼠股四頭肌Ⅰ型肌纖維比例較正常對(duì)照組明顯降低,相應(yīng)的Ⅱ型肌纖維比例明顯升高(P0.05)。 4.COPD模型組大鼠股四頭肌較正常對(duì)照組TWEAK表達(dá)明顯升高,PGC-1α表達(dá)明顯降低(P0.05)。 5.各組大鼠股四頭肌中TWEAK的表達(dá)與骨骼�、裥屠w維的比例的相關(guān)性分析呈負(fù)相關(guān),而與Ⅱ型纖維比例呈正相關(guān)。 6.各組大鼠股四頭肌中PGC-1α的表達(dá)與骨骼�、裥屠w維的比例的相關(guān)性分析呈正相關(guān),而與Ⅱ型纖維比例呈負(fù)相關(guān)。 7.各組大鼠股四頭肌中TWEAK的表達(dá)與PGC-1α的表達(dá)呈負(fù)相關(guān)。 結(jié)論： 1.COPD模型鼠股四頭肌Ⅰ型纖維明顯減少,Ⅱ型纖維明顯增多。 2.TWEAK的表達(dá)增加可能是COPD模型鼠股四頭肌中肌纖維類型發(fā)生轉(zhuǎn)換原因之一。 3. COPD模型鼠中,TWEAK可能是通過抑制PGC-1α的表達(dá),從而引起骨骼肌纖維類型轉(zhuǎn)換。
[Abstract]:Objective: to determine the proportion of various types of fibers in quadriceps femoris of rats with chronic obstructive pulmonary disease (COPD).And the expression of TWEAKA, a weak inducer of tumor necrosis factor-like apoptosis, and Y co-stimulator of peroxisome proliferation-activated receptor Y (PGC-1 偽) in the quadriceps femoris of COPD rats.To explore whether the increase of TWEAK expression in quadriceps femoris of COPD rats resulted in the decrease of PGC-1 偽 expression and the further change of fiber types of quadriceps femoris.Methods: fifty Wistar rats were randomly divided into control group (n = 20) and Wistar model group (n = 30).Lung function and general condition of rats were used to judge whether the model was successful or not.Rat quadriceps femoris were weighed and made into frozen sections.
