【摘要】：目的觀察IL-13對(duì)9HTE氣道上皮細(xì)胞內(nèi)Egr-1、Bax、Bcl-2 m RNA和蛋白表達(dá)的影響,探討IL-13在氣道上皮細(xì)胞凋亡中的作用及機(jī)制。方法將9HTE氣道上皮細(xì)胞分為正常對(duì)照組、rh IL-13組及rh IL-13+m AIL-13組,用逆轉(zhuǎn)錄-多聚酶鏈反應(yīng)(RT-PCR)法檢測(cè)Egr-1、Bax、Bcl-2 m RNA表達(dá)水平,用免疫細(xì)胞化學(xué)(ICC)法檢測(cè)Egr-1、Bax、Bcl-2蛋白表達(dá)水平。結(jié)果rh IL-13組Egr-1、Bax m RNA及蛋白表達(dá)顯著高于正常對(duì)照組(P0.01),Bcl-2 m RNA及蛋白表達(dá)顯著低于正常對(duì)照組(P0.01);rh IL-13+m AIL-13組Egr-1、Bax m RNA及蛋白表達(dá)顯著低于rh IL-13組(P0.01),與正常對(duì)照組比較無(wú)顯著性差異(P0.05),Bcl-2 m RNA及蛋白表達(dá)顯著高于rh IL-13組(P0.01),與正常對(duì)照組比較無(wú)顯著性差異(P0.05)。結(jié)論 IL-13可能通過(guò)Egr-1促進(jìn)9HTE氣道上皮細(xì)胞凋亡;m AIL-13可以中和IL-13所致的9HTE氣道上皮細(xì)胞凋亡的作用。
[Abstract]:Objective to observe the effect of IL-13 on the expression of Bcl-2 m RNA and protein in 9HTE airway epithelial cells, and to explore the role and mechanism of IL-13 in the apoptosis of airway epithelial cells. Methods 9HTE airway epithelial cells were divided into two groups: normal control group, rh IL-13 group and rh IL-13 m AIL-13 group. The expression of Bcl-2 m RNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results the expression of Egr-1Bax-Bax RNA and protein in rh IL-13 group was significantly higher than that in normal control group (P 0.01), Bcl-2 m RNA and protein expression were significantly lower than that in normal control group P0.01 IL-13 m AIL-13 and Egr-1 Bax m RNA and protein table. The expression of Bcl-2 m RNA and protein in rh IL-13 group was significantly higher than that in rh IL-13 group, and there was no significant difference between Rh IL-13 group and normal control group in the expression of Bcl-2 m RNA and protein. Conclusion IL-13 may promote 9HTE airway epithelium through Egr-1. Apoptotic AIL-13 can neutralize the apoptosis of 9HTE airway epithelial cells induced by IL-13.
