細(xì)菌溶解產(chǎn)物對(duì)哮喘小鼠氣道炎癥的預(yù)防作用

【摘要】:目的探討細(xì)菌溶解產(chǎn)物(OM-85BV)對(duì)哮喘氣道炎癥的預(yù)防作用及可能機(jī)制。方法 4~6周齡BALB/c小鼠48只,隨機(jī)分為6組。a組,空白對(duì)照組;b組,OM-85BV對(duì)照組;c組,哮喘模型組;d組,地塞米松(Dex)干預(yù)組;e組,OM-85BV干預(yù)A組;f組,OM-85BV干預(yù)B組(干預(yù)時(shí)間較e組延長(zhǎng)10 d)。在實(shí)驗(yàn)第1、8和15天分別給c、d、e、f4組小鼠腹腔注射卵清白蛋白(OVA)-Al(OH)3懸液使其致敏,其余組以PBS緩沖液對(duì)照。第17~26天,OM-85BV溶于生理鹽水(NS)并給f組灌胃,其余5組以生理鹽水(NS)灌胃。第27~31天分別給c、d、e和f組OVA的NS溶液滴鼻激發(fā),其余兩組以等量NS對(duì)照。其中b、e和f組在激發(fā)前給予OM-85BV灌胃,d組給予Dex灌胃,其余組用NS對(duì)照。末次激發(fā)后24 h麻醉處死小鼠,心臟采血測(cè)定血清總IgE水平,左肺行肺泡灌洗留取肺泡灌洗液(BALF)進(jìn)行細(xì)胞計(jì)數(shù)并計(jì)算嗜酸性粒細(xì)胞(Eos)比例,測(cè)定BALF上清液中IFN-γ和IL-4水平,取部分右肺行蘇木精-伊紅(HE)染色作病理學(xué)檢查。結(jié)果與a、b組比較,c組小鼠支氣管痙攣收縮明顯,支氣管上皮增生紊亂,支氣管周圍可見(jiàn)以淋巴細(xì)胞和嗜酸性細(xì)胞為主的炎癥細(xì)胞浸潤(rùn),管腔內(nèi)有少量黏液和脫落的上皮細(xì)胞,而且肺泡間隔增寬;BALF中炎性細(xì)胞總數(shù)、Eos比例增高,BALF上清中IL-4水平增高,而IFN-γ水平降低,血清中總IgE水平增高,差異均有統(tǒng)計(jì)學(xué)意義(P0.01);經(jīng)干預(yù)的d、f組較c組情況明顯改善,e組次之,以上各項(xiàng)指標(biāo)的差異均有統(tǒng)計(jì)學(xué)意義(P0.01)。結(jié)論口服OM-85BV干預(yù)有利于調(diào)節(jié)Th1/Th2之間的平衡,減輕小鼠哮喘模型氣道炎癥,可有效預(yù)防支氣管哮喘氣道炎癥的發(fā)生和發(fā)展。
[Abstract]:Objective to investigate the preventive effect and possible mechanism of bacterial dissolution product (OM-85 BV) on airway inflammation of asthma.On the 8th and 15th day of the experiment, the mice in the control group were sensitized with ovalbumin ovalbumin ovalbumin (OVA) -Albumin OHH3 suspension respectively, and the other groups were sensitized with PBS buffer solution.OM-85BV was dissolved in NS on the 26th day of 17 ~ (th) and the other 5 groups were given intragastric perfusion with NS (n = 5).On the 31st day of 27 ~ (th) day, the NS solution of OVA in group C and F were given nasal drip respectively, and the other two groups were treated with the same amount of NS as control group.Group B and group f were given OM-85BV intragastric perfusion before stimulation, and group B and F were given Dex before stimulation. The rest group were treated with NS as control group.The mice were anesthetized 24 hours after the last stimulation. The serum total IgE level was measured in the heart. The cell count and eosin ratio were calculated in the alveolar lavage fluid (BALF) of the left lung, and the levels of IFN- 緯 and IL-4 in the supernatant of BALF were measured.Results compared with the control group, the bronchial spasm was obvious, the bronchial epithelium was dysplastic, the inflammatory cells were mainly infiltrated by lymphocytes and eosinophils around the bronchus, and a small amount of mucus and exfoliated epithelial cells were found in the lumen.In addition, the ratio of total inflammatory cells to Eos was increased in BALF, while the level of IL-4 in supernatant of BALF was increased, the level of IFN- 緯 was decreased, and the level of total IgE in serum was increased.The difference was statistically significant (P 0.01), and that of the intervention group was significantly better than that of group C (P 0.01), and the difference of the above indexes was statistically significant (P 0.01).Conclusion Oral OM-85BV intervention can regulate the balance between Th1/Th2 and reduce airway inflammation in mouse asthma model. It can effectively prevent the occurrence and development of bronchial inflammation in asthma.
