COPD患者肺泡及外周血單核源性巨噬細(xì)胞Cofilin的表達(dá)與其吞噬煙曲霉功能的相關(guān)性

【摘要】:目的探討慢性阻塞性肺疾病(COPD)患者肺泡及外周血單核源性巨噬細(xì)胞Cofilin的表達(dá)與其吞噬煙曲霉功能的相關(guān)性。方法選取2015年7月至2016年5月南方醫(yī)科大學(xué)珠江醫(yī)院呼吸內(nèi)科門診20名COPD患者,其中觀察組1(2011版GOLD中A和B組),觀察組2(2011版GOLD中C和D組),健康對(duì)照組10人。3組行支氣管肺泡灌洗術(shù)、采取外周血,分別提取AM和MDM,檢測(cè)各組的AM和MDM對(duì)煙曲霉孢子的吞噬能力。運(yùn)用實(shí)時(shí)熒光定量(qRT-PCR)和免疫蛋白印跡實(shí)驗(yàn)(Western blot)檢測(cè)3組Cofilin蛋白的表達(dá),比較其差異。結(jié)果 3組人群MDM/AM吞噬煙曲霉孢子的菌落數(shù)分別為(17±3)個(gè)、(16±2)個(gè)、(42±3)個(gè)(F=73.446,P0.001),兩觀察組與健康組差異明顯,而兩觀察組無明顯差異。qRT-PCR和Western blot結(jié)果顯示,兩觀察組中Cofilin蛋白表達(dá)明顯高于健康組。結(jié)論 COPD患者肺泡及外周血單核源性巨噬細(xì)胞吞噬煙曲霉功能降低可能與Cofilin蛋白升高有關(guān)。
[Abstract]:Objective to investigate the relationship between the expression of Cofilin in alveolar and peripheral blood mononuclear macrophages and the phagocytic function of Aspergillus fumigatus in patients with chronic obstructive pulmonary disease (COPD). Methods from July 2015 to May 2016, the Pearl River Hospital of Southern Medical University was selected. Twenty patients with COPD in respiratory medicine outpatient department, Group A and B of GOLD were treated with bronchoalveolar lavage, group C and group D were treated with bronchoalveolar lavage and peripheral blood was treated with peripheral blood. The phagocytic ability of AM and MDM to Aspergillus fumigatus spores was determined by AM and MDM, and the expression of Cofilin protein was detected by real-time fluorescence quantitative analysis and Western blotanalysis. Results the colony number of MDM/AM phagocytic Aspergillus fumigatus spores in the three groups was 17 鹵3), 16 鹵2) and 42 鹵3 respectively. The difference between the two groups was significant, but there was no significant difference between the two groups. QRT-PCR and Western blot showed that there was no significant difference between the two groups. Conclusion the decrease of pulmonary alveolar and peripheral blood mononuclear macrophages phagocytosis of Aspergillus fumigatus in patients with COPD may be related to the increase of Cofilin protein.
