染矽塵大鼠結(jié)締組織生長因子和Ⅱ型肺泡細(xì)胞表面抗原的表達(dá)

【摘要】:[目的]探討結(jié)締組織生長因子(CTGF)與Ⅱ型肺泡細(xì)胞表面抗原(KL-6)在染矽塵大鼠血清、支氣管肺泡灌洗液(BALF)及肺組織中的表達(dá)及意義。[方法]將49只Wistar雄性大鼠隨機(jī)分為對照組和染塵后7、14、21、28、60、90 d共7個(gè)組,每組7只,染塵組采用非氣管暴露法灌注二氧化硅懸浮液(100 mg/m L),對照組以同樣方法灌注生理鹽水,取其肺組織并采集血清及BALF。采用免疫組織化學(xué)法測定肺組織中CTGF和KL-6的表達(dá)水平;采用酶聯(lián)免疫吸附法(ELISA)檢測血清及BALF中CTGF、KL-6的質(zhì)量濃度。[結(jié)果]免疫組化結(jié)果顯示,染塵大鼠肺組織中CTGF和KL-6呈陽性表達(dá),且染塵組的平均光密度值均高于對照組,差異有統(tǒng)計(jì)學(xué)意義(P0.05);染塵后隨時(shí)間的延長,CTGF表達(dá)呈先增后降的趨勢,在染塵后28 d達(dá)到最高峰,差異有統(tǒng)計(jì)學(xué)意義(P0.05);KL-6呈持續(xù)上升趨勢,且差異有統(tǒng)計(jì)學(xué)意義(P0.05)。ELISA結(jié)果顯示,染塵大鼠血清及BALF中CTGF質(zhì)量濃度的變化趨勢基本與其在肺組織中的一致,但是在染塵后21 d達(dá)到峰值;KL-6在血清及BALF中的變化也基本與肺組織中一致。[結(jié)論]CTGF和KL-6與肺纖維化有密切的關(guān)系,在矽肺形成過程中發(fā)揮了作用。
[Abstract]:[Objective] to investigate the relationship between CTGF and KL-6 in serum of rats exposed to silica dust. Expression and significance of BALF in bronchoalveolar lavage fluid and lung tissue. [Methods] Forty-nine male Wistar rats were randomly divided into 7 groups, 7 in each group. In the dust-infected group, the silica suspension was perfused with 100 mg/m / L of silica suspension by non-tracheal exposure, while the control group was perfused with normal saline in the same way. The expression of CTGF and KL-6 in lung tissue was determined by immunohistochemical method. Enzyme linked immunosorbent assay (Elisa) was used to detect the concentration of CTGFKL-6 in serum and BALF. [Results: the positive expression of CTGF and KL-6 was found in the lung tissues of the dust-exposed rats. The average optical density of the dust-exposed rats was higher than that of the control group (P 0.05). The expression of CTGF increased first and then decreased with time, and reached its peak at 28 days after dust exposure, the difference being statistically significant (P 0.05). KL-6 showed an increasing trend, and the difference was statistically significant (P 0.05). Elisa results showed that. The change trend of CTGF concentration in serum and BALF of dust-exposed rats was basically the same as that in lung tissue, but reached the peak at 21 days after dust exposure. The changes of KL-6 in serum and BALF were similar to those in lung tissue. [Conclusion CTGF and KL-6 are closely related to pulmonary fibrosis and play an important role in silicosis formation.
