鈣離子介導(dǎo)的卵母細(xì)胞凍融性損傷的初步研究

卵母細(xì)胞冷凍復(fù)蘇后的復(fù)蘇率、受精率、胚胎發(fā)育率和成活率一直不理想,導(dǎo)致這種現(xiàn)象的原因比較復(fù)雜,其中胞內(nèi)鈣離子濃度的升高可能是發(fā)育率降低的一個(gè)重要原因。為了探究細(xì)胞內(nèi)鈣離子的改變對(duì)卵母細(xì)胞造成的影響,本研究通過透射電子顯微鏡、激光掃描共聚焦顯微鏡等對(duì)冷凍前后各組卵母細(xì)胞的超微結(jié)構(gòu)、精子穿透、透明帶消化、發(fā)育率以及鈣離子濃度和波動(dòng)進(jìn)行研究。結(jié)果發(fā)現(xiàn):(1)透射電子顯微鏡下,新鮮對(duì)照組、毒胡蘿卜素(Thapsigargin,TG)處理組、2-氨基乙氧基苯硼酸(2-Aminoethoxydiphenylborate,2-APB)處理組卵母細(xì)胞的透明帶和線粒體結(jié)構(gòu)都比較完整;而冷凍組、TG冷凍組卵母細(xì)胞的冷凍損傷較嚴(yán)重,透明帶變寬,邊緣處破損,卵周隙變寬,線粒體結(jié)構(gòu)模糊不清;2-APB冷凍組卵母細(xì)胞損傷較輕,透明帶和線粒體等細(xì)胞器結(jié)構(gòu)只有部分出現(xiàn)模糊。(2)透明帶消化時(shí)間和透明帶結(jié)合精子個(gè)數(shù):2-APB冷凍組(230.5±7.14,4.17±1.11)、TG 冷凍組(172.5±5.91,1.17± 1.03)、冷凍組(221.22±9.46,2.75±1.29)與新鮮對(duì)照組(300.14±7....
