【摘要】：Cyclin D1和Cyclin E1均為細(xì)胞G1/S期關(guān)鍵蛋白。本研究旨在克隆牦牛(Bos grunniens)Cyclin D1和Cyclin E1基因的CDS序列,分析其生物學(xué)特性,并研究胰島素樣生長(zhǎng)因子-I(Insulin-like growth factor-I,IGF-I)和轉(zhuǎn)化生長(zhǎng)因子-β1(Transforming growth factor-β1,TGF-β1)對(duì)Cyclin D1在睪丸支持細(xì)胞(Sertoli cell,SC)中表達(dá)的影響。采用RT-PCR技術(shù)克隆牦牛Cyclin D1和Cyclin E1基因,采用免疫熒光染色技術(shù)對(duì)其蛋白在SCs中的表達(dá)進(jìn)行了定位分析。采用實(shí)時(shí)定量PCR(qRT-PCR)和蛋白免疫印跡法檢測(cè)不同濃度的IGF-I(0(對(duì)照組)、25、50、100、150 ng?m L~(-1))和不同時(shí)間(0(對(duì)照組)、5、12、18、24 h)、不同濃度(0、250、500、750、1000 pg?mL~(-1))的TGF-β1對(duì)Cyclin D1基因和蛋白表達(dá)的影響。結(jié)果如下:(1)牦牛Cyclin D1和Cyclin E1基因與其它物種的同源性均較高,GenBank登錄號(hào)分別為KY 420723和KY437813。其蛋白在SCs胞核中均高表達(dá),為核蛋白。(2)IGF-I濃度為25和150 ng?mL~(-1)作用組Cyclin D1 mRNA表達(dá)量與對(duì)照組(0 ng?m L~(-1))差異不顯著(P0.05),50和100 ng?m L~(-1)作用組Cyclin D1 mRNA表達(dá)量顯著高于其他各組(P0.05);100 ng?mL~(-1)作用組Cyclin D1蛋白表達(dá)量與對(duì)照組差異不顯著(P0.05),25、50和150 ng?mL~(-1)作用組Cyclin D1蛋白表達(dá)量顯著高于對(duì)照組(P0.05);IGF-I濃度為50 ng?m L~(-1)時(shí),Cyclin D1 mRNA和蛋白表達(dá)量均為最高,分別為對(duì)照組的1.65和1.20倍。(3)TGF-β1作用于SCs 5、12和24 h時(shí),Cyclin D1 mRNA的表達(dá)量顯著高于對(duì)照組(P0.05),作用到18 h時(shí),其表達(dá)量顯著低于其它組(P0.05);不同時(shí)間處理組的Cyclin D1蛋白表達(dá)量都顯著高于對(duì)照組(P0.05)。TGF-β1各濃度處理組中Cyclin D1 mRNA和蛋白表達(dá)量均顯著高于對(duì)照組(P0.05)。TGF-β1以時(shí)間和劑量依賴的方式促進(jìn)Cyclin D1的表達(dá),最佳時(shí)間和濃度分別為12 h和750 pg?m L~(-1)。結(jié)果表明,Cyclin D1和Cyclin E1在進(jìn)化過程中高度保守,其蛋白質(zhì)定位于細(xì)胞核;IGF-I和TGF-β1可以通過促進(jìn)Cyclin D1的表達(dá)來調(diào)控SCs的生長(zhǎng)。本研究為調(diào)控SCs的發(fā)育提供了實(shí)驗(yàn)依據(jù)。

2.2 CyclinD1、CyclinE1 基因 CDS 序列克隆及生物信息學(xué)分析Cyclin D1-C、Cyclin E1-C 進(jìn)行 RT-PCR 擴(kuò)增后，瓊脂糖凝膠電泳顯示，預(yù)期位置有明顯熒光條帶（圖 2）。利用 NCBI 在線軟件 BLAST 對(duì)拼接后的序列進(jìn)圖 1 細(xì)胞培養(yǎng)和鑒定（標(biāo)尺 A~D：100 μm，，E：50μm）Fig.1 The cultivation and identification of the cells (Bar: A~D:100 μm,E:50μm)A. 原代 SCs； B. 三代 SCs；C.
