錳中毒導(dǎo)致雞免疫器官免疫和炎性損傷的研究

【摘要】:錳(Manganese,Mn)是自然界中廣泛分布的一種元素,錳在生命活動(dòng)的不同環(huán)節(jié)都表現(xiàn)出生物學(xué)功能,與酶的合成、三大物質(zhì)代謝、造血、免疫和生殖功能有直接的關(guān)系。錳是機(jī)體必不可少營養(yǎng)元素,但過量錳對(duì)生物體有害。錳對(duì)神經(jīng)、生殖、免疫系統(tǒng)造成的損傷已被證實(shí)。但錳對(duì)雞免疫系統(tǒng)細(xì)胞因子和炎癥因子影響的研究還很少見。本研究通過建立雞錳中毒模型,探討錳中毒對(duì)雞免疫器官細(xì)胞因子和炎癥因子的影響。7日齡180只海藍(lán)蛋公雞分為4組,對(duì)照組飼喂標(biāo)準(zhǔn)飼料,3個(gè)處理組飼喂標(biāo)準(zhǔn)飼料分別添加不同濃度的Mn Cl2(飼料中含錳的濃度分別為600、900和1800 mg/kg)。在錳處理30、60和90天后,分別采集法氏囊,胸腺和脾臟,測(cè)定三個(gè)免疫器官中IL-4、IL-7、IL-10、IL-17、TGF-β4、IFN-γ、NF-κB、TNF-α、COX-2、PGE2和iNOSm RNA表達(dá)。采用單因素方差分析方法,分析了不同組間檢測(cè)因子的差異性,并運(yùn)用多元相關(guān)分析、主成分分析、聚類分析三個(gè)方法分析檢測(cè)的細(xì)胞因子之間的關(guān)系,試驗(yàn)得出如下結(jié)論:1.30天和60天錳中毒上調(diào)了法氏囊、胸腺和脾臟NO含量、iNOS活力、IL-4、IL-7、IL-10、IL-17、TGF-β4、NF-κB、TNF-α、COX-2、PGE2和iNOS mRNA表達(dá),抑制了IFN-γm RNA表達(dá),90天錳中毒抑制了以上所有檢測(cè)因子,過量錳導(dǎo)致形態(tài)學(xué)的免疫損傷和炎癥損傷。60天胸腺IL-7、60天脾臟NF-κB、30天的胸腺iNOS和30天脾臟PGE2的m RNA表達(dá)隨著時(shí)間的增加而增加。2.IL-4、IL-7、IL-10和IL-17的關(guān)系較近,且與IFN-γm RNA表達(dá)成負(fù)相關(guān)關(guān)系。3.錳中毒導(dǎo)致了雞法氏囊、胸腺和脾臟免疫損傷和炎性損傷。
[Abstract]:Manganese (Manganese,Mn) is a widely distributed element in nature. Manganese shows biological function in different links of life activities, which is directly related to enzyme synthesis, three major substance metabolism, hematopoiesis, immunity and reproductive function. Manganese is an essential nutrient element in organism, but excessive manganese is harmful to organism. Damage to the nervous, reproductive and immune systems caused by manganese has been confirmed. However, the effects of manganese on immune system cytokines and inflammatory factors in chickens are rarely studied. In this study, the effects of manganese poisoning on cytokines and inflammatory factors in immune organs of chickens were studied by establishing a model of manganese poisoning. 180 7-day-old sea blue egg roosters were divided into 4 groups and the control group was fed with standard diet. Three treatment groups were fed with standard diet supplemented with different concentrations of Mn Cl2 (feed manganese concentrations were 600900 and 1800 mg/kg, respectively). After manganese treatment for 30, 60 and 90 days, bursa of Fabricius, thymic gland and spleen were collected to detect the expression of IL-4,IL-7,IL-10,IL-17,TGF- 尾 4, IFN- 緯, NF- 魏 B, TNF- 偽, COX-2,PGE2 and iNOSm RNA in three immune organs. The difference of detection factors among different groups was analyzed by single factor variance analysis, and the relationship among cytokines was analyzed by multivariate correlation analysis, principal component analysis and cluster analysis. The results are as follows: 1.30 and 60 days manganese poisoning upregulated NO content, iNOS activity, IL-4,IL-7,IL-10,IL-17,TGF- 尾 4, NF- 魏 B, TNF- 偽, COX-2, in bursa of Fabricius, thymus and spleen. The expression of PGE2 and iNOS mRNA inhibited the expression of IFN- 緯 mRNA, manganese poisoning inhibited all the above factors on the 90th day, excessive manganese induced morphological immune injury and inflammatory injury. On the 60th day of thymic IL-7,60, spleen NF- kappa B, 2. Il-4, IL-7, IL-10 and IL-17 were closely related to the expression of m-RNA in thymus iNOS and 30-day spleen PGE2. 3. The expression of IL-4, IL-7, IL-10 and IL-17 was negatively correlated with the expression of IFN- 緯 m RNA. Manganese poisoning results in immune and inflammatory damage to bursa Fabricius thymus and spleen in chickens.
