獲得最大酵母多糖提取率的破壁方法研究

【摘要】:試驗(yàn)旨在選擇獲得最大酵母多糖提取率的酵母菌破壁方法。以酵母多糖提取率與細(xì)胞破壁率為綜合評(píng)定指標(biāo),首先對(duì)超聲波破碎、超聲波清洗、反復(fù)凍融、蛋白酶處理4種方法進(jìn)行條件優(yōu)化,選擇最適破壁條件;然后將此4種單一的破壁方法與超聲波破碎和蛋白酶處理協(xié)同方法相比較,確定獲得最大多糖提取率的破壁方法。結(jié)果表明,當(dāng)超聲波清洗時(shí)間達(dá)到40min、超聲波破碎時(shí)間達(dá)到50min時(shí),酵母多糖提取率達(dá)到最大值;當(dāng)菌泥加水量和凍融次數(shù)分別為15%與3次時(shí),多糖提取率最大;當(dāng)?shù)鞍酌赣昧?00IU/g、酶解時(shí)間15h、pH 5.0、處理溫度40℃時(shí),蛋白酶處理的多糖提取率最高。4種破壁方法中蛋白酶處理的效果最佳,對(duì)應(yīng)的多糖提取率為149.81mg/g,破壁率為53.49%。協(xié)同破壁處理的提取效果要優(yōu)于單一的破壁方法,但考慮到協(xié)同作用對(duì)多糖結(jié)構(gòu)的完整性構(gòu)成威脅,將蛋白酶處理確定為最佳的酵母菌破壁方法。
[Abstract]:The purpose of the experiment was to select a method for obtaining the maximum extraction rate of yeast polysaccharide by yeast broken wall. Taking the extraction rate of yeast polysaccharide and the rate of cell breaking as the comprehensive evaluation index, the conditions of ultrasonic crushing, ultrasonic cleaning, repeated freezing and thawing, and protease treatment were optimized, and the optimum conditions of wall breaking were selected. Then the four single wall breaking methods were compared with the ultrasonic crushing and protease treatment methods to determine the maximum extraction rate of polysaccharide broken wall method. The results showed that when the ultrasonic cleaning time reached 40 min and the ultrasonic crushing time reached 50min, the extraction rate of yeast polysaccharide reached the maximum, when the amount of water added to the sludge and freeze-thaw times were 15% and 3 times, respectively, the extraction rate of polysaccharide was the highest.
