【摘要】：奶牛乳腺炎是造成奶牛飼養(yǎng)場(chǎng)經(jīng)濟(jì)損失最大的一種常見(jiàn)疾病,因此如何有效的防治奶牛乳房炎已經(jīng)越來(lái)越受到奶牛飼養(yǎng)企業(yè)的重視。一直以來(lái),防治奶牛乳房炎的方法大多采用抗生素,隨之帶來(lái)的耐藥性和藥物殘留等問(wèn)題漸漸暴露出來(lái),中藥防治以它無(wú)污染、天然和無(wú)毒的特性進(jìn)入人們的視線。本研究選取了幾味中藥,通過(guò)體外抑菌試驗(yàn)確立了最優(yōu)的提取方法并配制了復(fù)方中藥乳房灌注劑,對(duì)配制好的復(fù)方中藥乳房灌注劑進(jìn)行了安全性評(píng)價(jià),并在臨床上進(jìn)行了治療試驗(yàn)。試驗(yàn)結(jié)果如下:1查閱文獻(xiàn)設(shè)計(jì)試驗(yàn),通過(guò)體外抑菌試驗(yàn)效果確定最優(yōu)的提取方法和條件:蒲公英、甘草和當(dāng)歸采用水煮法合并濃縮得到中藥提取液,百里香通過(guò)揮發(fā)油提取器回流提取百里香揮發(fā)油,將提取的中草藥均勻混合。體外抑菌和MIC的效果說(shuō)明配制的中藥乳房灌注劑對(duì)乳房炎的四種常見(jiàn)病原菌有良好的抑制效果。在臨床醫(yī)治奶牛乳房炎上提供了理論和實(shí)驗(yàn)基礎(chǔ)。2為了評(píng)價(jià)自制的中藥復(fù)方乳房灌注劑的安全性,對(duì)其進(jìn)行了粘膜刺激試驗(yàn)與急性毒性試驗(yàn),粘膜刺激試驗(yàn)結(jié)果表示本復(fù)方乳房灌注劑對(duì)粘膜無(wú)刺激性,經(jīng)口灌服給藥時(shí),半數(shù)致死量LD50=31063mg/kg,該劑量遠(yuǎn)小于藥物學(xué)急性毒性試驗(yàn)規(guī)定的標(biāo)準(zhǔn),所以認(rèn)為該藥物安全無(wú)毒,由此可證實(shí)本試驗(yàn)復(fù)方中藥乳房灌注劑劑對(duì)小鼠安全無(wú)毒,可在臨床上安全使用。3試驗(yàn)選擇乳房炎奶牛進(jìn)行分組,分為對(duì)照組、中藥灌注劑組和抗生素組進(jìn)行臨床治療試驗(yàn),觀察試驗(yàn)?zāi)膛５呐R床癥狀、體溫、體細(xì)胞數(shù)和血清生化指標(biāo)的變化,得出以下結(jié)果:中藥乳房灌注劑對(duì)乳房炎有一定的治療效果,替代抗生素使用可以克服抗生素易產(chǎn)生耐藥性、藥物殘留等缺點(diǎn)。
[Abstract]:Cow mastitis is a common disease which causes the biggest economic loss in dairy cattle farm. Therefore, how to effectively prevent and cure cow mastitis has been paid more and more attention by dairy cattle breeding enterprises. The methods of preventing and curing cow mastitis have been mostly used antibiotics, and the problems of drug resistance and drug residue have been exposed gradually. Traditional Chinese medicine has been used to prevent and cure dairy cow mastitis because of its pollution-free, natural and non-toxic characteristics. In this study, several kinds of traditional Chinese medicine were selected, the optimum extraction method was established by in vitro bacteriostasis test, and the breast perfusion agent of compound Chinese medicine was prepared, and the safety of the prepared Chinese herbal medicine breast perfusion agent was evaluated. A therapeutic trial was carried out in clinical practice. The results are as follows: (1) according to the literature design experiment, the optimal extraction method and conditions were determined by in vitro bacteriostasis test: dandelion, Glycyrrhiza and Angelica sinensis were condensed to obtain traditional Chinese medicine extract by water boiling method. The volatile oil of thyme was extracted by reflux of thyme oil extractor, and the extracted Chinese herbal medicine was mixed evenly. The results of in vitro bacteriostasis and MIC showed that the prepared Chinese medicine breast perfusion had a good inhibitory effect on the four common pathogens of mastitis. In order to evaluate the safety of traditional Chinese medicine compound breast perfusion agent, mucosal stimulation test and acute toxicity test were carried out. The results of mucosal stimulation test showed that this compound mammary perfusion agent had no irritation to the mucous membrane. When administered orally, the lethal dose of LD50=31063mg/kg, was far less than the standard prescribed in the pharmacological acute toxicity test, so the drug was considered safe and non-toxic. It can be proved that this compound Chinese medicine breast perfusion agent is safe and non-toxic to mice, and can be safely used in clinic. 3 the experiment selected mastitis cows to be divided into control group, traditional Chinese medicine perfusion group and antibiotic group for clinical treatment. By observing the changes of clinical symptoms, body temperature, somatic cell count and serum biochemical indexes of dairy cows, the following results were obtained: Chinese medicine breast perfusion has a certain therapeutic effect on mastitis. Alternative antibiotic use can overcome the disadvantages of antibiotic resistance and drug residue.
