新城疫弱毒活疫苗中FAdV-4的分離鑒定及分子特征
[Abstract]:[objective] at present, the avian adenovirus (Fowl adenovirus,FAd V) infection is serious in China. The virus can be transmitted both horizontally and vertically through eggs, so whether there is FAd V contamination in live vaccine of chicken embryo source poultry. The spread of the disease, in turn, is unclear. To detect the presence of FAd V pollution in commercial Newcastle disease (NDV) La Sota attenuated live vaccine in China. To explore the possible causes of FAd V infection and epidemic in domestic chickens. [methods] the commercial Newcastle disease La Sota attenuated vaccine was detected by FAd V PCR method established in our laboratory, and the FAd V was isolated and identified. By sequence alignment and evolutionary analysis, we identified the species, serotype and evolutionary characteristics of the isolated strain. [results] A avian adenovirus strain named L160962 was isolated. Gene analysis showed that the sequence of 52k and hexon gene fragments of strain L160962 had the highest similarity with serotype 4 of strain C, and the similarity with HN/151025 and CH/HNJZ/2015 of Chinese strain was 99.9- 100. The similarity with the Austrian strain KR5 was 99.3, but the similarity with FAd V was 79.0-83.3, respectively. Phylogenetic analysis showed that L160962 had the same branch with serotype 4 adenovirus, and the nucleotide similarity was more than 99.9%, which should be divided into serotype 4 FAd V. Based on the topological structure of phylogenetic tree constructed by hexon gene of isolate L160962, the phylogenetic relationship between L160962 and Chinese strain was close. The results suggest that the vaccine source strain L160962 is close to that of the current epidemic strain in China. [conclusion] serotype 4 FAd V contamination exists in the commercial Newcastle disease La Sota attenuated live vaccine in China. This may be one of the causes of FAd V infection and prevalence in Chinese chickens.
