杜仲葉對綿羊脂肪代謝的影響及其機(jī)理

【摘要】:為研究杜仲葉對綿羊脂肪代謝和肉品質(zhì)的影響,本研究隨機(jī)選擇70~80日齡、體重25~30 kg的綿羊(湖羊)30只,平均分為3組,分別為對照組(CTL組,飼糧中不含杜仲葉)、低水平杜仲葉組(EUL1組,飼糧含10%杜仲葉)和高水平杜仲葉組(EUL2組,飼糧含20%杜仲葉),每組10只。預(yù)試期15 d,正試期90 d。采集血液,測定血漿脂質(zhì)代謝分析指標(biāo);采集背最長肌,測定營養(yǎng)物質(zhì)含量;采集肝臟組織,測定脂肪代謝相關(guān)酶和核轉(zhuǎn)錄因子的mRNA表達(dá)水平和蛋白水平。結(jié)果表明:1)各組干物質(zhì)采食量EUL1組EUL2組CTL組,組間差異顯著(P0.05)。2)與CTL組相比,EUL2組血漿極低密度脂蛋白(VLDL)、高密度脂蛋白(HDL)、非酯化脂肪酸(NEFA)含量顯著升高(P0.05),EUL1和EUL2組血漿低密度脂蛋白(LDL)(0.05≤P0.10)、總膽固醇(TC)(P0.05)含量降低。3)與CTL組相比,EUL1組肌肉粗蛋白質(zhì)含量、EUL2組肌肉粗脂肪含量顯著升高(P0.05),EUL1組肌肉剪切力顯著減小(P0.05)。4)與CTL組相比,EUL1組和EUL2組肌肉飽和脂肪酸(SFA)含量略有降低(P0.05),肌肉不飽和脂肪酸(UFA)含量顯著升高(P0.05),其中,EUL2組肌肉MUFA、PUFA含量及UFA/SFA顯著升高(P0.05)。5)與CTL組相比,杜仲葉能夠顯著下調(diào)肝臟脂肪合成相關(guān)酶硬脂酰輔酶A去飽和酶1(SCD1)和核轉(zhuǎn)錄因子固醇調(diào)節(jié)元件結(jié)合蛋白1c(SREBP-1c)、過氧化物酶體增生激活受體γ(PPARγ)的mRNA表達(dá)水平(P0.05);顯著上調(diào)肝臟脂肪酸分解相關(guān)酶肉毒堿棕櫚;D(zhuǎn)移酶1A(CPT1A)、脂蛋白酯酶(LPL)mRNA表達(dá)水平顯著上調(diào)(P0.05);與CTL組相比,EUL2組肝臟SCD1蛋白水平顯著下降(P0.05)。結(jié)果提示,杜仲葉通過調(diào)節(jié)脂肪合成和分解相關(guān)基因的表達(dá),顯著影響綿羊脂肪的代謝和肌肉脂肪酸的組成。
[Abstract]:In order to study the effect of Eucommia ulmoides leaves on fat metabolism and meat quality of sheep, 30 sheep (Hu sheep) were randomly divided into three groups (CTL group, diet without Eucommia ulmoides). Low level Eucommia ulmoides leaves group (EUL1 group, diet containing 10% Eucommia ulmoides leaves) and high level Eucommia ulmoides leaf group (EUL2 group, diet containing 20% Eucommia ulmoides leaves), 10 rats in each group. The pre-test period was 15 days and the current test period was 90 days. Blood was collected and plasma lipid metabolism was measured; longissimus dorsi muscle was collected to determine the content of nutrients; liver tissue was collected to determine the mRNA expression level and protein level of lipid-metabolization-related enzymes and nuclear transcription factors. The results showed that: 1) there was significant difference between CTL group and CTL group in EUL1 group (P0.05). 2. Compared with CTL group, very low density lipoprotein (VLDL), high density lipoprotein (HDL), in EUL2 group was higher than that in CTL group.
