禽波氏桿菌山東地區(qū)不同分離株的致病性研究

【摘要】:為檢測(cè)禽波氏桿菌雞胚分離株和呼吸道分離株對(duì)雛雞的致病性差異,本研究對(duì)2004年~2011年分離鑒定的12株禽波氏桿菌進(jìn)行了雞胚半數(shù)致死量和雛雞半數(shù)致死量的檢測(cè),篩選強(qiáng)毒菌株進(jìn)行雛雞的鼻腔感染試驗(yàn)。結(jié)果表明LL、XT和AQ分離株的毒力最強(qiáng),其它分離株的毒力稍弱。3株強(qiáng)毒菌株均可感染雛雞呼吸道和肺臟,引起呼吸道癥狀以XT和AQ的癥狀最為明顯,而在其它內(nèi)臟中造成感染的雞只比例以LL株最高,可達(dá)30%~40%;AQ最低,僅10%。HE染色結(jié)果顯示對(duì)氣管的損傷以XT最強(qiáng),而肺臟、肝臟等內(nèi)臟器官的病理變化以LL最嚴(yán)重。免疫酶組化試驗(yàn)也證實(shí)LL在內(nèi)臟組織中的載量要多于XT和AQ。3株菌株均可導(dǎo)致雛雞的生長(zhǎng)抑制和免疫器官損傷,對(duì)新城疫滅活苗免疫后的抗體水平具有輕微的抑制作用,且均以LL的抑制作用最強(qiáng)。實(shí)驗(yàn)結(jié)果顯示在所檢測(cè)的12株禽波氏桿菌中,雞胚分離株LL毒力最強(qiáng),具有比呼吸道分離株XT和AQ更強(qiáng)的感染雛雞內(nèi)臟組織的能力,本研究為進(jìn)一步鑒定禽波氏桿菌毒力因子和免疫原性奠定了基礎(chǔ)。
[Abstract]:In order to detect the pathogenicity of chicken embryo isolates and respiratory tract isolates of Boxella fowls to chicks, the 50% lethal dose of chicken embryos and the 50% lethal dose of chicken chickens were determined in 12 isolates isolated from 2004 to 2011. The nasal cavity infection test of chicks was carried out by screening virulent strains. The results showed that the virulence of LL,XT and AQ isolates was the strongest, while the virulence of other isolates was weaker. All three virulent strains could infect the respiratory tract and lung of chicks, and the respiratory symptoms of XT and AQ were the most obvious. In other viscera, the proportion of chickens infected with LL strain was the highest, up to 30% and 40%. AQ was the lowest, only 10%.HE staining showed that XT was the most serious injury to trachea, while LL was the most serious in visceral organs such as lung and liver. The immunohistochemical test also confirmed that LL in visceral tissue was more than that of XT and AQ.3 strains, which could induce growth inhibition and immune organ damage in chicks, and had a slight inhibitory effect on the antibody level of Newcastle disease inactivated vaccine after immunization. The inhibitory effect of LL was the strongest. The results showed that the chicken embryo isolate (LL) was the most virulent of the 12 strains detected, and had stronger ability to infect chicken visceral tissue than respiratory isolates XT and AQ. This study laid a foundation for further identification of virulence factor and immunogenicity of B. fowls.
