【摘要】：為了分離鑒定寧夏某肉牛場牛支原體和分析該病原對靶器官的損傷,采用Thiaucourt's液體篩選培養(yǎng)基和固體培養(yǎng)基進行病原分離,設(shè)計牛支原體16SrRNA通用引物和uvrC特異性引物進行基因序列擴增并測序,使用DNA Star軟件將分離菌株測序結(jié)果與GenBank中的標準株序列進行同源性比較,采用MEGA6.0軟件中的鄰接法(Neighbor-joining,NJ)依據(jù)16SrRNA和uvrC序列構(gòu)建分離株系統(tǒng)發(fā)育樹并進行遺傳進化分析,采用石蠟切片,HE染色,病理組織學觀察。結(jié)果表明,病原菌落呈油煎蛋樣,16S rRNA和uvrC擴增片段與陽性對照一致,16S rRNA和uvrC基因序列構(gòu)建的系統(tǒng)發(fā)育樹與牛支原體在同一分支;肺組織結(jié)構(gòu)消失,部分肺泡腔實變、塌陷,局灶性肺出血,肺泡腔內(nèi)可見炎性細胞浸潤;肺泡隔增厚、出血,肺泡內(nèi)有少量脫落的上皮細胞;肺泡壁斷裂,肺泡腔可見紅色的炎性滲出物,有纖維結(jié)締組織增生。
[Abstract]:In order to isolate and identify Mycoplasma bovis in a beef farm in Ningxia and analyze the damage of the pathogen to the target organ, Thiaucourt's liquid screening medium and solid medium were used to isolate the pathogen. 16SrRNA universal primers and uvrC specific primers for Mycoplasma bovis were designed to amplify the gene sequence and sequenced. The sequence of isolated strains was compared with that of standard strains in GenBank by DNA Star software. The phylogenetic tree of isolated strains was constructed by Neighbor-joining,NJ in MEGA6.0 software according to 16SrRNA and uvrC sequences, and the phylogenetic analysis was carried out. Paraffin sections, HE staining and histopathology were used to observe the phylogenetic tree. The results showed that the pathogenic bacteria were like fried eggs, the 16s rRNA and uvrC amplified fragments were consistent with the positive control, and the phylogenetic tree constructed by 16s rRNA and uvrC gene sequence was in the same branch as Mycoplasma bovis. The pulmonary tissue structure disappeared, partial alveolar cavity consolidation, collapse, focal pulmonary hemorrhage, inflammatory cell infiltration in alveolar cavity, alveolar septum thickening, hemorrhage, a small number of exfoliated epithelial cells in the alveoli. Alveolar wall rupture, alveolar cavity visible red inflammatory exudates, fibrous connective tissue proliferation.
