應(yīng)用Illumina-MiSeq高通量測序技術(shù)分析脫氧雪腐鐮刀菌烯醇對小鼠腸道菌群的影響

【摘要】:本試驗(yàn)旨在研究脫氧雪腐鐮刀菌烯醇(DON)對小鼠腸道微生物多樣性、物種豐度的影響。選用平均體重為20 g的BALB/c小鼠40只,隨機(jī)分成對照組(灌胃滅菌生理鹽水)和DON組(灌胃1.8 mg/kg BW DON),每組20個(gè)重復(fù),每個(gè)重復(fù)1只小鼠,連續(xù)灌服28 d。試驗(yàn)結(jié)束后,收集新鮮糞便,每組隨機(jī)選取3份,用Illumina-Mi Seq高通量測序技術(shù)分析微生物群落結(jié)構(gòu)和組成的變化。結(jié)果表明,1)與對照組相比,DON組測序數(shù)量減少(P0.05)。2)DON組降低了Alpha、Beta多樣性,Shannon指數(shù)較對照組顯著降低(P0.05)。3)在門水平,與對照組相比,DON組顯著降低了擬桿菌門(Bacteroidetes)和脫鐵桿菌門(Deferribacteres)的物種豐度(P0.05),顯著提高了變形菌門(Proteobacteria)的物種豐度(P0.05)。在屬水平,與對照組相比,DON組顯著降低了副類桿菌屬(Parabacteroides)、理研菌屬(Rikenella)、Algoriphagus、Mucispirillum、嗜甲基菌屬(Methylophilus)、Francisella的物種豐度(P0.05),顯著提高了梭菌屬(Clostridium)、Robinsoniella、A llobaculum、A kkermansia的物種豐度(P0.05)。4)聚類分析顯示,DON組與對照組的腸道微生物相似性降低。由此可見,DON能顯著影響小鼠腸道微生物菌群多樣性及物種豐度,提示DON致腸道損傷與這些腸道菌群的變化密切相關(guān)。
[Abstract]:The purpose of this study was to study the effects of deoxynivalenol (DON) on the microbial diversity and species abundance in the intestinal tract of mice. Forty BALB/c mice with an average body weight of 20 g were randomly divided into two groups: control group (normal saline by gastric administration) and DON group (20 replicates in each group of 1.8 mg/kg BW DON), by gavage for 28 days). At the end of the experiment, fresh feces were collected and three samples were randomly selected from each group. The changes of microbial community structure and composition were analyzed by Illumina-Mi Seq high-throughput sequencing technique. The results showed that: 1) compared with the control group, the number of DON sequencing decreased (P0.05). The DON group decreased the Alpha,Beta diversity, and the Shannon index significantly decreased compared with the control group (P0.05). DON group significantly decreased the species abundance of (Bacteroidetes) and (Deferribacteres) (P0.05), and significantly increased the species abundance of (Proteobacteria) (P0.05). At the generic level, compared with the control group, the DON group significantly decreased the species abundance of the genus (Parabacteroides), Rhinopsis, (Rikenella), Algoriphagus,Mucispirillum, (Methylophilus), Francisella (P0.05), and significantly increased the species abundance of the genus (Clostridium), Robinsoniella,. Cluster analysis of species abundance of A llobaculum,A kkermansia (P0.05) showed that the similarity of intestinal microbes between DON group and control group was decreased. It can be seen that DON can significantly affect the diversity and species abundance of intestinal microflora in mice, suggesting that the intestinal injury induced by DON is closely related to the changes of these intestinal flora.
